ObjectiveTo investigate the VDR gene Taq I sites, Pepsinogen C(PGC) and TP53PIN3gene polymorphism's distribution of the Han and Dai Nationality in Yunnan Sporadic brea-st cancer (SBC) and non-breast cancer population. To analyze the association between the three genes'polymorphism and the susceptibility to SBC in Han and Dai people, and compare with the three genes'polymorphism which have ethnic difference or not.Method(1) Collect blood specimens of200patients with SBC in Yunnan, contain80Dai cases and120Han cases, while in the control group of200patients: Dai80cases and Han120cases.(2) VDR gene Taq I sites, PGC and TP53PIN3genotypes' distribution features were detected by polymerase chain reaction-restriction fragment length poly-morphism (PCR-RFLP) in four group.(3) Chi-square test was adopted to investigate the difference in genotype distribution and check the accordance with Hardy-Weinbe-rg equilibrium between patients and the controls group, and analyze the risk between TP53PIN3polymorphism and breast cancer.Result(1) The distribution of VDR genotypes in Han and Dai people were in line with Hardy-Weinberg genetic equilibrium equation (PH-w>0.05). The frequencies of TT, Tt, tt of VDR gene Taq I sites were35.0%,52.5%,12.5%and37.5%,53.7%,8.8%in Han and Dai cases group respectively, There were no significant difference (χ2=0.711, P=0.701). The incidence of TT, Tt, tt was no significance between the Han and Dai natio-nality cases and controls group (χ2=0.523, P=0.770; χ2=0.270, P=0.874).(2) PGC allele2,3,4,5in Han and Dai cases and the controls group were no statist-ical significance (P>0.05), PGC alleles1(310bp) in the Han, Dai nationality cases and the control group was no statistical significance (χ2=0.356, P=0.551; χ2=0.417, P=0.519), but the positive rate in Han, Dai cases group were32/120(26.67%),34/80(42.50%), a significant difference was found in two nationality (χ2=5.442, P=0.020).(3) The frequencies of TP53genotypes AA and AA" were68.3%,31.7%in Han SB C group and85%,15%in the control group respectively, there was significant differ-ence (χ2=9.317,P=0.002); The frequencies were66.3%,33.8%in Dai SBC group and82.5%,17.5%in the control group respectively, there was significant difference (x2=5.542, P=0.019). But no statistical difference was found in Han and Dai's cases group (P>0.05).(4) Risk analysis was used for estimations of OR values for breast cancer, showing that TP53PIN3OR were2.62(95%CI:1.396-4.939) and2.402(95%CI:1.146-5.033) in Han, Dai cases and the control group.Conclusion(1)VDR gene Taq I polymorphism was not associated with the susceptibility in Han, Dai people with sporadic breast cancer.(2) PGC allele polymorphism was not related with Han, Dai sporadic breast cancer susceptibility. But the distribution of PGC allele1polymorphism was different between Han and Dai nationality cases group, showing that exist inter-ethnic differences between Han and Dai nationality.(3) There was asso-ciation between TP5316bp insertion polymorphism and Han, Dai nationality, while there were no ethnic differences between two group.(4) TP53gene mutation AA" was the risk factor in sporadic breast cancer, may increase the risk of sporadic breast can-cer in Han and Dai nationality people.
