Objective To investigate the relationship between Genetic Polymorphisms of Vitamin D receptor,Pepsinogen C,TP53 and the susceptibility of Breast Cancer in Yunnan women of han nationality. Methods VDR,PGC and TP53gene polymorphisms were detected using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis, and the genotypes were identified. VDR,PGC and TP53gene polymor-phisms were obtained from a Yunnan population-based case-control study including 80 sporadic breast cancer and 80 normal controls. Results The VDR genotypes identified were TT, Tt, tt; the TP53 genotypes identified were A1A1, A1A2, A2A2; the PGC gene was detected in 5 alleles,15 geno-types. VDR gene TT, Tt, tt frequency distribution of three genotypes, breast cancer group were 36.3%,48.6% and 15.0% in the control group were 38.6%,46.3%,15.0% VDR genotype distribution between the two groups showed no significant difference (X2=0.119, P=0.942>0.05); Breast cancer patients in T,t allele frequencies were 60.6%,39.4%, in the control group were 61.8%,38.2%, VDR allele frequen-cies between the two groups showed no significant difference (X2=0.053 P=0.818>0.05)。TP53 gene A1A1, A1A2, A2A2 frequency distribution of three genotypes in breast cancer group were 52.5%,30.0% and 17.5% in the control group were 66.3%,32.3% and 2.5%,TP53genotype distribution between the two groups showed significant difference (X2=10.294 p=0.006); breast cancer patients Al, A2 allele frequency was 67.5%,32.5%,in the control group were 81.9%, 18.1%, TP53 allele frequencies between the two groups showed significant difference (X2=8.744,P=0.003), PGC allele inthe breast cancer group were 31.9%,27.5%, 20.0%,13.1%,6.7%, in the control group were 29.3%,28.1%,20.6%,13.8%,8.1%, PGC allele (Z=-0.201,P=0.841>0.05) and genotype distribution frequency between the two groups showed no significant difference (X2=0.253, P=0.993>0.05)。Conclusion TP53 gene polymorphism is associated with sporadic breast cancer in Yunnan; VDR gene polymorphism TaqI is not associated with sporadic breast cancer in Yunnan; PGC gene polymorphism is not associated with sporadic breast cancer in Yunnan. Genotype A2A2 TP53 might be susceptible genotype in sporadic breast cancer in Yunnan, A2 allele might be the susceptibility gene in sporadic breast cancer in Yunnan.
