Mappianthus iodoies Hand. MaZZ., which is the general medical materials named TongZuan of Chinese Yao medical, has some quite special efficacy. On the contrary with the various researchs on its components such as the chemical compounds of ketones, terpenes and alkaloids, the research on the polysaccharides component is limited. As a kind of bioactivator, the polysaccharides hold an important medical position thus is widely used in the medical domain. Besides, Mappianthus iodoies Hand. MaZZ. is a kind herbal medicine derived from the minority of south China, and additional to the strong ethnic characteristics, it also possesses historical traditional medical theory valuable for research and reference. Consequently, implementing research on the polysaccharides of Mappianthus iodoies Hand. MaZZ. will comprehensive benefit displaying the application worth.The study mainly included the best extracted conditions of Water-soluble Polysaccharides from Mappianthus iodoies Hand. MaZZ.(DXTc), determination of physical and chemical properties of DXTc, isolation and purification of DXTc, all compositions of DXT, DXT1, DXT2,DXT3, DXT4, DXT5, structure determination of DXT51and free radical scavenging activity of DXT, DXT1, DXT2, DXT3,DXT4, DXT5 and DXT51. The results can provide scientific theoretical basis for understanding the pharmacological effects and application of Mappianthus iodoies Hand. MaZZ.. The experimental results are as follows:Orthogonal experiment analysis:extraction conditions of DXTc were liquid ratio1:8, extraction temperature80℃, extracting time2h, the extraction rate is1.6%.The respective contents for protein, polysaccharides and uronic acid of HMc were9.91%,65.66%and15%.DXT was produced by hydrolysis starch and deproteinization of DXTc. Acidic ethanol classification of DXT to get DXT1, DXT2, DXT3, DXT4, DXT5. DXT5was purified by DEAE-SephadexA-25and SephadexG-15, to get DXT51. HPLC showed a single symmetrical and PC showed a single spot, which indicate DXT51was purified polysaccharides. Its molecular weight is about20000.PC and GC analysis indicated that DXT51is composed of rhamnose(Rha), arabinose(Ara), xylose(Xyl), mannose(Man), galactose(Gal), glucose(Glc), and not containing galacturonic acid. Monosaccharide composition of molar ratio is1.0:10.9:0.3:1.2:2.6:6.2.In addition to the GC analysis, IR, GC, Periodate oxidation, Smith degradation, Methylation GC-MS and NMR method were also used to analyze the structure of DXT51. The results showed that DXT51is branching structure, its main chain is made up of Ara and Glc. One of main chain is a-(1â†'4) Ara or a-(1â†'5) Ara connect with a-(1â†'2) Ara, another is a-(1â†'6)Glc with branches on C4, and an average of5branch among13residues. The side chains were composed of (1â†'3)Rha,(1â†'3)Ara,(1â†'3)Man,(1â†'2)Gal and (1â†'3)Glc. The terminal residues were composed of Rha, Ara, Xyl, Gal and Glc.The free radical scavenging experiment showed that DXT, DXT,, DXT2, DXT3, DXT4, DXT5and DXT51with the ability to scavenge OH and DPPH, and the clearance rate is correlated with the concentration of polysaccharide and reaction time, that means the polysaccharides of Mappianthus iodoies Hand. MaZZ. is a good scavenging agent for OH and DPPH.
