The traditional Yao's folk-medicine, which named Mappianthus iodoies is a plant of Mappianthus Hand.-Mazz. which belongs to Icacinaceae. It distributes in our country north west to south various provincial areas. Its roots has the efficacy of dispelling wind and dampness and relieving pain, and can regulate menstruation and make the blood circulation. It is used for to treat rheumatism arthritis, kind of rheumatic arthritis, the jaundice, the injury from fractures, the abnormal menstruation, the painful menstruation, the menopause, it is very useful for governs the flesh wound hemorrhage, the poisonous snake bites. Its chemical constituents have been reported less, thus the extraction, separation and structural identification of Mappianthus iodoies were carried out. The results showed that it contains flavonoids, so we make the research of extraction and purification of total flavonoids from Mappianthus iodoies. These results reassured rattan for further efficacy of research and development resources to provide certain scientific basis.The 75% EtOH extract of Mappianthus iodoies was re-extracted with petroleum ether, chloroform, ethyl acetate, and water successively to give four fractions, among of which, fraction petroleum ether, chloroform and ethyl acetate were isolated and purified by utilizing column chromatograph, TLC techniques, recrystallization, and twelve compounds were yielded and ten compounds were identified by IR, MS, 1H NMR, 13C NMR and traditional phytochemical methods. It including taraxerone(1), Tricosanoic acid(2),β-sitosterol(3), Daucosterol(4), Vanillin(5), Chrysin(6), Vanillic acid(7), Quercetin(8), Gallic acid(9), Sinomenine(10). All compounds were first isolated from Mappianthus iodoies.The contents of total flavonoids was determined by UV spectrophotometrer. Orthogonal experiment was carried out. The influence of concentration of sovelt, the dosage of solvent,time length of extraction, and frequency of extraction on extraction results were investigated by using the content of pueraria as inex. The best extraction conditions are to use 65%(V/V) ethanol as extractor with the radio of 1:10 at the temperature of 80℃for 2.5h, and extracting 3 times. Under this condition, by means of D-101 macroporous resin, the crude flavonoids extraction was purified . The content of total flavonoids in the refined product was 86%.
