Objective: Primary bronchogenic carcinoma (or lung cancer), refers tothe malignant tumor whose origin is the bronchial mucosa and gland. Lungcancer is generally divided into two categories: non-small cell lung cancer(NSCLC), small cell lung cancer (SCLC), where NSCLC accounted formore than80%of all lung cancer. NSCLC is the most commonly malignanttumor, whose prognosis is very poor in the world, and the rate of5yearsurvival is less than15%. In recent years, with the development of molecularbiology, looking for NSCLC specific tumor markers to improve diagnosticrate and molecular targeted therapy to improve treatment efficiency havebecome the key to diagnosis and treatment. Inhibiting cancer cell apoptosisand tumor angiogenesis are the main reason in the development of NSCLC,whose mechanism is complex, involving many related genes and proteins. Theinactivation and deficiency of suppressor genes and activation of oncogeneshave been considered to be important events in the development of NSCLC.Inhibitor of DNA binding-3(Id-3) is a cancer gene, which can inhibit celldifferentiation and promote cell proliferation by the regulation of the cell cycle.It plays an important role in inducing tumor occurrence, promoting tumorangiogenesis and tumor invasion and regulation. The current study hasdiscovered that the Id-3is over-expression in cancer cells, but its research inNSCLC is less. PTEN discovered in1997is a tumor suppressor gene, whichplays an important role in inhibition of tumor cell proliferation, differentiation,tumor invasion and metastasis and tumor angiogenesis. This paper study theexpression of Id-3and PTEN in NSCLC,and analyze their effect in thegeneration and development of NSCLC.Methods:134paraffin-embedded specimens were collected and thespecimens were got from the patients with NSCLC in the second chest surgery department in the fourth affiliated hospital of Hebei Medical University from2007.6to2008.6. The pathological results were all confirmed by thedepartment of pathology of the fourth affiliated hospital of Hebei MedicalUniversity.36normal tissues (operated tissues from benign lung disease) wereconsidered as the control group. The cases were separately grouped accordingto the age, gender, sizes of tumor, differentiation, pathological type,clinicalstage of tumor, lymph node metastasis etc. We applied semi-quantitativeimmunohistochemistry method to detecting the expression of Id-3and PTEN.All specimens are observed in the medical image analysis system of MoticMED6digital. The expression of Id-3stains in the cytoplasm, yellow tobrown granules; the expression of PTEN is coloring in the cell nucleus, yellowor brown granules. We used the method of staining intensity and thepercentage of staining cells to analyze the results. We had done the contrastanalysis to find out the reasons of the different expression of Id-3and PTEN ineach group and the correlation analysis to find out the relationship of Id-3andPTEN in all NSCLC cases. We used Spearman correlation analysis to analyzethe correlation between the expression of Id-3, PTEN. P <0.05indicates thatthe difference has statistics significance. We consider P<0.05as statisticallydifference.Results:1Id-3positive expression rate in NSCLC tissues and normal tissues is76.1%(102/134),11.1%(4/36)respectively. The deviation is statistical significance(P <0.05).2Id-3expression does not correlate with patients' sex, age, tumor size andpathological type (P>0.05), has a correlation with differentiation, lymph nodemetastasis and the clinical stage (P <0.05).3PTEN positive expression rate in NSCLC tissues and normal tissues is44.0%(59/134),97.2%(35/36),The deviation is statistical significance(P<0.05).4PTEN expression does not correlate with patients' sex, age, tumor size andpathological type (P>0.05), has a correlation with differentiation, lymph node metastasis and the clinical stage (P <0.05).5The expression of Id-3and PTEN is negatively correlated (r=-0.420, P=0.007).Conclusions:1Id-3expression rate in NSCLC tissues is higher than this in normal tissues,implying that Id-3over-expression is relevant to NSCLC.2The expression of Id-3is relevant to the degree of malignancy and tumorinvasion and metastasis. The higher the expression of Id-3is, the lower tissuedifferentiation of NSCLC is, the more likely lymph node metastase, the laterclinical stage is.3The expression of PTEN in NSCLC rate is lower than this in normal tissues,inferring that PTEN low-expression is relevant to NSCLC.4The expression of PTEN is relevant to the degree of malignancy and tumorinvasion and metastasis. The lower the expression of PTEN is, the lower tissuedifferentiation of NSCLC is, the more likely lymph node metastase, the laterclinical stage is.5Id-3and PTEN are negatively relevant in the occurrence and metastasis andinvasion of NSCLC, implying that Id-3activation and inactivation ordeficiency of PTEN promote the occurrence and development of NSCLC.
