Abstract/Summary:
Objective: Epilepsy is a chronic disease of nervous system causedby variety of reasons. Seizure is resulting form electro-discharge of neuron.There are many women of childbearing age suffer from Epilepsy. Pregnanciesoccurring in women who are epileptic are considered to be high risk. Allwomen with epilepsy who are of childbearing age should be advised(preferably before conception) that the incidence of malformations in infantsof mothers with epilepsy who are treated with antiepileptic drugs (AEDs) istwo or three times that of infants of mothers without epilepsy. Themalformations are caused by the genes of the parents, and the use of AEDs.Uncontrolled epilepsy is also associated with maternal and fetal risk. Tonicclonic seizure may affect fetal development by intrauterine asphyxia andtrauma. With the development of society, science and technology, people aremore and more interested in health. In addition to no diseases andmalformations, healthy children have normal development, normalpsychology and normal behavior. Seizuring during pregnancy as a harmfulstimulus, may lead to deficiency of development. p38is a MAPK(Mitogen-activated protein kinase) that has been shown to induce a widevariety of biological effects in cell culture in response to a wide range ofstimuli. In this research, we establish chronic epileptic pregnant modelinducing by pentylenetetrazol (PTZ). Then we observed the dates ofdevelopment and measure the expression of p38and P-p38protein inhippocampus of fetal rat.Methods: Chronic epileptic models were induced by intraperitonealinjection of pentylenetetrazol (PTZ,35mg/kg).36female SD rats (7-8weeks,body weight200±20g) were randomly divided into normal control group (NC group), saline group (NS group) and the epilepsy group (PTZ group).Each group has12rats. From the first day, rats in the PTZ group were injectedPTZ intraperitoneally by35mg/kg at9:00every day, the rat in NS group wereintraperitoneally injected with the same doses of saline, the NC groupreceived no treatment. We observed one hour after each injection. Accordingto Racine criterion, if we observed three times successive seizures of grade Ⅳor Ⅴ in3days, the kindling in epileptic rats was successful. Then we give thetreatment another week. Rats will instead by others if they die or fail to bekindled. After the female rat was kindled, female rat were mated with males.Pregnancy was confirmed by the presence of a vaginal plug. If there hasvaginal plug, we will make the day as0days of gestation. And every groupcontinue to pre-pregnancy treatment until20days pregnant. In each group wewould select randomly6rats, and take out the the fetal rat by caesareansection with anesthetization of chloralic hydras. The expression of p38andP-p38protein in hippocampus of fetal rat will be measured by the methods ofWestern-Blot. Keep the rest of pregnant females until delivery. At birth, thefollowing data were recorded: the litter sizes, number of live and dead pups,individual pup length, individual pup weight on PND1,2,4,7and14.The dayof occurrence for pinna detachment, incisor eruption, eye opening, rightingreflex on PND2to6, grip response on PNG3to7were also monitored.Results:1Results of physical developmentThere was no significant difference of litter sizes, number of live anddead pups among NC,NS and PTZ group(P>0.05). Individual pup length,weight on PND1,2,4,7and14was no significant difference among NC,NSand PTZ group(P>0.05). The day of occurrence for pinna detachment,incisor eruption, eye opening was no significant difference among NC,NS andPTZ group(P>0.05). Righting reflex on PND2to5was significant differenceamong NC,NS and PTZ group(P<0.05). Righting reflex on PND6was nosignificant difference among NC,NS and PTZ group(P>0.05). Grip responseon PND3to5of PTZ group was delayed obviously compared both to NC and NS group (P<0.05). There was no significant deference between NC groupand NS group(P>0.05). Grip response on PND6-7was no significantdifference among NC,NS and PTZ group(P>0.05).2Results of Western blot:P38in hippocampus of fetal rat was no significant difference amongNC,NS and PTZ group(P>0.05). P-P38in hippocampus of PTZ group wasincreased obviously compared both to NC and NS group (P<0.05). There wasno significant deference between NC group and NS group(P>0.05).Conclusions:1Seizuring during pregnancy may not affect litter sizes, number of liveand dead pups, individual pup length, weight on PND1,2,4,7and14, the dayof occurrence for pinna detachment, incisor eruption, eye opening of pups,but delay the reflex development.2As a noxious stimuli, seizuring during pregnancy may induces thephosphorylation of P38, and Increased expression of P-P38. All of thesemay further activate the apoptotic pathway which delay the reflexdevelopment.
