The Expression Of GAP-43and Sema3A In Hippocampus Of The Kindled Rats

Objective This study was investigated the expression of GAP-43(growth-associated protein43) and Sema3A(semaphoring3A) in the hippocampus of epileptic rats.Methods Pentylenetetrazol(PTZ,40mg/kg) were used to establish chronic epileptic models by intraperitoneal injection.41rats were divided into saline treated control group(NC group) and epilepstic group(EP group) randomly. The immunohistochemistry and western blot was used to detect the expression of GAP-43and Sema3A. in hippocampus.Results1. There were four rats dead in EP group. The success rate of establishment model was81%, and the mortality rate was19%.2. Immunohistochemistry results: Compared with the NC group, the number of GAP-43positive cells in the CA1,CA3and DG(Dentate Gyrus) of the hippocampus was significantly increased in EP group (P<0.05). In the CA1of hippocampus, Sema3A positive cells in EP group was significantly higher than those in NC group (P<0.01). Meanwhile, in the CA3and DG of hippocampus, Sema3A positive cells in EP group was significantly lower than those in NC group(P<0.01).3. Western blot results:Expression of GAP-43protein in hippocampus of EP group(0.45±0.21)was significantly higher than those in NC group(0.28±0.23)(P<0.05). Expression of Sema3A protein in hippocampus of EP group(1.18±0.55)was lower than those in NC group’s (1.55±1.96), but there was no statistically significant difference(P>0.05).Conclusion Expression of GAP-43protein in hippocampus of EP group was higher. Expression of Sema3A was varied in different areas. The changes of the expression of GAP-43and Sema3A may be involved in the pathogenesis of epilepsy.
