Purpose:To study the chemical constituents, especially the steroidal saponins of the herb of Solanum lyratum Thunb, and then focusing on the separation of different parts and resulting compands of it for anti-tumor screening activity.Method:Compounds, especially the steroidal saponins, were isolated from the herb of Solanum lyratum Thunb by column chromatography on macroporous resin860021, silica-gel and octadecylsilanized silica-gel (ODS), and semi-preparative RP-HPLC. Their structures were determined by chemical evidence and1D NMR (1H NMR and13C NMR),2D NMR ('H-'H COSY, HSQC and HMBC) and MS spectra. Taking MTT method to observe the resulting compands'inhibition effect on human gastric carcinoma cell SGC-7901.Results:Fourteen compounds were isolated and determined as (25R)-5(6)-en-spirost-3β-ol-3-O-β-D-glucopyranosyl-(1â†'4)-[α-L-rhmanopyranosyl-(1â†'2)]-β-D-galactopyranoside (1), β-sitosterol (2),5,7-diol-8-methoxy-flavone (3),(25R)-5a-spirost-3β-ol-3-O-β-D-glucopy-ranosyl-(1â†'2)-β-D-glucopyranosyl-(1â†'4)-β-D-galactopyranoside (4),(25R)-5(6)-en-spirost3β-ol-3-O-β-D-glucopyranosyl-(1â†'2)-β-D-glucopyranosyl-(1â†'4)-β-D-galactopyranoside (5),16,23-epoxy-22,26-epimino-cholest-22(N),23,25-trien-3β-ol-3-O-β-D-glucopyranosyl-(1â†'2)-β-D-glucopyranosyl-(1â†'6)-β-D-galactopyranoside (6),(25R)-5α-spirost-3β-ol-3-O-β-D-glucopyranosyl-(1â†'3)-[β-D-glucopyranosyl-(1â†'2)]-β-D-glucopyranosyl-(1â†'4)-β-D-galactopyranoside (7), Diosgenin (8), Tigogenin (9),26-O-β-D-glucopyranosyl-(25R)-5(6),20(22)-dien-furost-3β26-diol (10),26-O-β-D-glucopyranosyl-(25R)-5a-furost-3β,26-diol (11),(25R)-5(6)-en-spirost-3β-ol-3-O-β-D-xylopyranosyl-(1â†'3)-[β-D-glucopyranosyl-(1â†'2)]-β-D-glucopyranosyl-(1â†'4)-β-D-galactopyranoside (12),(25R)-5α-spirost-3β-ol-3-O-β-D-xylopyranosyl-(1â†'3)-[β-D-glucopyranosyl-(1â†'2)]-β-D-glucopyranosyl-(1â†'4)-β-D-galactopyranoside (13), Ursolic acid (14). The alcohol extract of95%ethanol-eluting solution of ethyl acetate extraction layer after sample has a obvious inhibitory effect on cell SGC-7901, with IC50=34.86μg/ml.Conclusion:Compounds6and11were new compounds. Compound4has a strong inhibition effect on human gastric carcionma cell SGC-7901, witth IC5o=13.90μg/ml. |
