Chemical Composition Of White English Extraction, Separation And Structural Identification

Solanum Lyratum Thunb. , a herb of Solanum, it also is called "baimaoteng" in China. Which is one of the most valued Chinese traditional medicine, has been used as herbal remedy for cancers, tumours and herpes over two centuries.Many researchers have studied the pharmacology , chemical constituents and clinical applications about Solanum Lyratum Thunb in the world. However, the data on its chemical composition and its biological activity are scarce. It is well known that extraction and purification of natural products from Chinese traditional medicine are not only key steps of rational combination, pharmacology and molecule structure identification, but also important for Chinese medicine development. So we didresearch on the chemical constituents about Solanum Lyratum Thunb.The chemical constituents of Solanaceae were studied by systematic solventextraction. The dried whole plant of solanum lyratum was extracted three times by ethanol (95%) , then partitioned with CH₃C1 and EtOAc and n-Bu-OH successively. During our chemical studies on this plant, 8 compounds were isolated from the n-Bu-OH extract part by sephadex LH-20 chromatography using CH₃OH, silica-column chromatography using (CH3CL- CH₃OH ) and thin-layer chromatography using (CH3CL- CH3OH -H2O) repeatedly, and 6 compounds were identified, their structures were identified on basis of physico-chemical evidences and spectra data. Their structures were determined as apigenin-7-0-β-D-apiofuanosyl(1â†'2)-β-D- glucopyranoside (I), apigenin- 7-O-β-D-glycopyranoside (II) ,adenosine (III) , 3-methoxy -4- hydroxy-5-[ (8'S) -3' -methoxy -4'- hydroxyl -phenylpropyl alcohol] -E-cinnamic- alcohol-4 -O-β- D- -glucopyranoside (IV) , N-(4-Amino-butyl) -3-(3-hydroxy-4 -methoxy -phenyl)-E- -acrylamide (V) , N-(4-Amino-butyl)-3- (3-hydroxy-4-methoxy-phenyl) -Z-acrylamide(VI). And these compounds were obtained for the first time from this genus.The ¹H and ¹³C-NMR of compound (V) was calculated theoretically at the levels B3LYP/6-311G+(2d, p) using the GIAO(gauge-independent atomic orbital) method.
