Purpose: In this study,Litchi seed was extracted, isolated and purified.Try to selecthypoglycemic activity site of Litchi seed Polysaccharide and determine physical and chemicalproperties and chemical structure.Method: By Water extraction and ethanol precipitation method polysaccharidefraction-LSEP was extracted from litch seed. LSEP were furthermore purified by D101macroporous resin; weak alkaline anion exchanger DEAE sephadex A-50; dextran gelSephadex G-50. UV spectrophotometry was carried out to determine total sugar and uronicacid content of different factions, HPLC analysis to calculate average molecular weightthrough Gel Permeation Phromatography (GPC) and determine monosaccharide compositionthrough PMP Derivatization Method. Fraction of higher Polysaccharide content:LSEP-2a-A,LSEP-2a-B,LSEP-2a-C,LSEP-2a-D was isolated and LSEP-2a-C will beproceeded structural analysis.Result: Main chain of LSEP-2a-C is characterized by1â†'4,1â†'6linkage Glc.In thebranch point, side chain was linked, which was composed of Man and Gal,and terminal wascomposed of Man.Conclusion: By above study,we extracted, isolated and purified hypoglycemic activitysite from litchi seed. Activity site was further purified and preliminarily identified chemicalstructure of litchi seed.
