Purpose: With inhibitory effect ofα-glucosidase as a pharmacodynamics index, glycoprotein from Marasmius androsaceus was screened, isolated and purified, physiochemical properties and chemical structure of which is characterized. We are trying to discover new activities of Marasmius androsaceus, and relevant active ingredient. Method: 80% Alcohol precipitation was conducted to purify MAEP from water extracts of fermentation product of Marasmius androsaceus. MAEP were further purified by hollow fiber super filter (MW 3000), weak alkaline anion exchanger DEAE sephadex A-50, agarose gel Sepharose CL-6B, dextran gel Sephadex G-100. UV spectrophotometry was carried out to characterize physiochemical properties of different factions, HPLC analysis to calculate average molecular weight and GC to determine monosaccharide composition through acid hydrolysis derivatives method. Meanwhile, inhibitory effects of different fractions onα-glucosidase were performed to track active fraction. Active fraction MAEP-2b-2a was isolated and subjected to structural investigation. Result: Main chain of MAEP-2b-2a is characterized by 1â†'2, 1â†'6 linkage. Mannose linked by 1â†'2 bonds has branch point in the fourth saccharide, while mannose linked by 1â†'6 bonds in the second one. In the branch point, side chain was linked, which was composed of Gal and Glu. Non-reducing end of MAEP-2b-2a comprises of Gal, Man and Glu. Conclusion: With progressive purification of MAEP, active fraction becoming centralized and total level of saccharide and protein rising, we deduce that glycoprotein from fermentation product of Marasmius androsaceus might be the active ingredient of inhibitory effect onα-glucosidase.
