Objective: To determine blood pressure and dipsogenic behavior in the offspring affected by high-salt diet during pregnancy.Methods:Pregnant Sprague Dawley rats were fed with high-salt food(8%NaCl)from gestation day 4 to 21 (high-salt SD rats, HSD); control rats were fed with normal food (1%NaCl) during gestion (normal-food SD rats, NSD). The offspring were fed with normal food. Blood pressure during base line period and after intracerebroventricular injection (i.c.v) of AngII was assessed. Blood gases and electrolytes were determined with a Nova Analyzer; water and sodium intake in the offspring was measured everyday for 14 days. Water and sodium intake of the adult offspring was tested after water deprivation for 24 hours or i.c.v of AngII.Results:Maternal high-salt diet during pregnancy did not chang the [Na+] and Osmolality(Osm) in the blood of adult offspring in base . The blood pressure was higher than that of the control rats after i.c.v injection of AngII. However, maternal high-salt diet during pregnancy significantly increased sodium intake. Whereas the water intake did not change. The [Na+] and Osm in the blood and water or sodium intake increased after dehydration for 24 hours and i.c.v injection of AngII.Conclusion: Maternal high-salt diet during pregnancy showed chronic influence on the offspring with the change of blood pressure and dipsogenic behavior. Objective: To investigate the impact on the brain RAS in the adult offspring following maternal high-salt diet pregnancy.Methods: The offspring in both the HSD and NSD groups were used. AngII concentration was measured by radioimmunoassay; MRNA and protein of ATR were measured with real-time PCR and western blot in both the forebrain and hindbrain of offspring. C-fos expression was detected after water deprivation for 24 hours and centrum AngII injection.Results:Compared to NSD, the plasma concentration of AngII in the HSD offspring was significantly reduced. Although it was increased after water deprivation for 24 hours. In the forebrain, AT2R mRNA was increased whereas AT1R mRNA did not change, however, AT1R and AT2R proteins were significantly increased. In the hindbrain, ever the AT1R and AT2R mRNA or protein did not change. However, the ratio of AT1R and AT2R for both mRNA and protein in the forebrain and hindbrain showed statistically significant difference. The c-Fos positive nuclei, induced by water deprivation and i.c.v injection of AngII, were obserived in the several concerned brain regions in the adult offspring.Conclusions: The results indicated that maternal high-salt diet affected the brain RAS and Ang II receptor expression patterns in the offspring.
