[Objectives] To investigate the impact on body homeostasis in the adult offspring affected by prenatal high sucrose diet.[Methods] Pregnant Sprague-Dawley rats were randomly divided into two groups. High sucrose (HS) group were given 20% sucrose solution and standard rat food from day 1 to day 21of gestation. Control group (C) was provided with fresh tap water and standard rat food. Blood electrolytes and plasma osmolity were determined using a Nova analyzer and an advanced digmatic osmometer in the adult offspring. Water intake and sodium appetite of the adult offspring were assessed after 24 hours water deprivation.[Results] Under basal condition, blood value and plasma osmolarity were not significantly different in HS adult offspring from that of the control. Blood hematocrit was significantly higher in the offspring of group HS than that of the control after 24 hours water deprivation. In additional, the HS adult offspring drank more water and saline (1.8% NaCl) than that of control after 24 hours water deprivation.[Conclusions] The"imprinting"effects may be linked to body fluid homeostasis probably affected by high sucrose diet during pregnancy.Part II Possible central mechanisms of fluid homeostasis in the adult offspring related to prenatal high sucrose[Objectives] To investigate the central mechanism in the centrol of fluid homeostasis in the adult offspring exposed to prenatal high sucrose.[Methods] Pregnant Sprague-Dawley rats were randomly divided into two groups. High sucrose (HS) group were given 20% sucrose solution and standard rat food from day 1 to day 21of gestation. Control group (C) was provided with fresh tap water and standard rat food. With the application of histological immunochemical staining, c-Fos positive staining in the concerned brain region ,such as the SFO, SON, PVN, OVLT, MnPO, AP and NTS were counted in the adult offspring following 24 hours water deprivation. The protein levels of AT1 and AT2 receptors were quantified by western-blot in the forebrain of the adult offspring.[Results] c-Fos positive cell in the concerned brain regions were significantly increased in the HS adult offspring compared to those in control. The expressions of AT1 and AT2 receptors in the forebrain of HS adult offspring were increased compared to that of the control.[Conclusions] The central mechanism for fluid homeostasis may be affected by prenatal high sucrose.
