The counteractive effects of the combination of LTP and ASA on gastric injury were investigated in this paper and the mechanisms of LTP on ASA-induced gastric injury were explored primarily. In the study, mice were intragastric administrated with the combination of LTP and ASA according to proportion of 25 mg·kg-1:100 mg·kg-1, 50 mg·kg-1:100 mg·kg-1 and 100 mg·kg-1:100 mg·kg-1. Rats were intragastric administrated according to proportion of 18.75 mg·kg-1:75 mg·kg-1, 37.5 mg·kg-1:75 mg·kg-1 and 75 mg·kg-1:75 mg·kg-1.The results showed that the combination of LTP and ASA 50:100 and 100:100 could lower the injury index of gastric mucosa in mice, and attenuate the aspirin-induced injury. Although each proportion had no effect on gastric juice and pepsin, the combination of 37.5:75 and 75:75 could significantly inhibit the H+ back-diffusion and increase free mucus in gastric juice in rats,.The malonaldehyde (MDA) level in both plasma and gastric mucosa was found to be reduced. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) in gastric mucosa in mice were increased. The combination of 50:100 and 100:100 could also increase NO level of serum and gastric mucosa, which had no significant effect on plasma ET, TXB2 and 6-keto-PGF. The combination of 37.5:75 and 75:75 could significantly increase GMBF detected by the neutral red clearance method in rats. Serum EGF level was enhanced remarkably by the combination of 75:75.In the study of relationship between blood concentration of LTP and pharmacodynamic action, plasma PGE2 level was significantly increased at the moment of 4.0 h and 12.0 h after administrated the combination of 75:75 in rats. The phenomenon of mucosal cellular abscission, necrosis and confused structure in rats decreased with the combination of 75:75 in each moment group. The results of the present investigation revealed that the combination of LTP and ASA had the anti-injury effects on ASA-induced gastric injury. In addition, the combination of LTP and ASA could inhibit pulmonary embolism induced by collagen and adrenaline in mice and platelet aggregation in vitro, which indicated that the combination of LTP and ASA had synergetic effects on anti-thrombosis and anti-aggregation.
