Animal development is a complex process that tissues, organs andorgan systems are generated gradually from a single-cell fertilized egg.Gametogenesis is an important initial event in the process of animaldevelopment. Gametogenesis, including spermatogenesis and oogenesis,gives rise to sperms and eggs. Gametogenesis is a complex biologicalprocess, which is dependent upon numerous factors, both intrinsic andextrinsic, as well as being species-specific.In previous studies we identified a mouse dynein axonemalintermediate chain2(Dnaic2) gene, which express a component ofaxonemal dynein complex that function in cilia or flagella. A series of invitro and in vivo experiments have revealed that Dnaic2has certainfunction in the process of reproductive development. In this study, weconstructed a Dnaic2knockdown mouse model for further investigationof the role of Dnaic2. PCR and Southern blot were used to screen thetransgenic mice, and reduced expressions of Dnaic2were confirmed by Western blot. We obtained36positive mice form126F1mice and thepositive rate was28.6%. Through analysis of the phenotype we foundthat55%transgenic mice died between50and56days after birth due tosevere lung inflammation and45%were infertilile or due to spermabnormality in male or oocyte abnormality and pathological changes oftunica mucosa in oviduct of female.In summary, our result of this study show that Dnaic2is essential formouse development and reduced expression of Dnaic2leads todysfunction in lung, ovary and testis. Our research provides severalfundamental clues for further study of the Dnaic2function.
