Structured lipids(SLs) is a particular lipid which modify or alter the coposition andlocation of the fatty acid on triacylglycerol, that is specific fatty acid residues located in a specificposition. The research of structured lipids is designed to optimize the fatty acidcomposition and arrangement to make use of each type of fatty acid functions andcharacteristics, which is a effective way to alleviate contradictory between variousdiseases cause by high energy dietary and use beneficial function of oil. Study theconditions of produce MLM-type(M, medium-chain fatty acid, L, long-chain fattyacid) structured lipids achieved by acidolysis of canola rapeseed oil and caprylic acidusing1,3-specific immobilized lipases in packed Based on single factors experiments,central composite design was adopted to investigate the influences of reaction time,substrate molar ratio, reaction temperature, water content and their interaction onproduce MLM-type structured lipids. Then the optimal conditions were obtained byresponse surface methodology.(1) Experimental results indicated that the best result was attained with reactiontime was2h, substrate molar ratio was4.5, reaction temperature was60℃and watercontent was0.15. Under these conditions the content of MLM-type structured lipidswas46.12. The established regression model shows high reliability and high degree offitting, which was feasible to predict yield.(2) The article research the methods of purification about KOH-hydroalcoholicsolution, hydroalcoholic solution and molecular distillation method, respectively,Experimental results indicated that the method of KOH-hydroalcoholic solution issimple, rapid and convenient in laboratory-scale. In the pilot scale moleculardistillation method have obvious advantage, with environmentally friendly and lessdamage of target product. the methods of purification abou combine KOH-hydroalcoholic solution with hydroalcoholic solution can reduce non-target generation and oil saponification in laboratory conditions, which have more obvious advantage inseparation and purification.(3) The MLM-type structured lipids produce in packed bed reactor is faintyellow liquid, have the structured lipids inherent odor and characteristic. In thesencory it is not many differences to substrate canola rapeseed, differences of moistureand volatile matter content, refractive index, melting point and acid value are small,the color is slightly darker than canola rapeseed oil. The iodine value, peroxide value,saponification value and oxidative stability are difference, gas chromatographyanalysis showed that the prepared MLM type structure in a lipid C8:0levels increasedsignificantly.
