In this study, the appropriate method of extraction of gliadin from pricot kernel was investigated by ethanol, ethylene glycol, isopropanol, tert-butanol and acetone, extraction rate was as the indicator. The effects of concentration and pH of organic solvents on extraction rate and conditions of isolating protein were investigated. Moreover, the effect of gliadin from pricot kernel extracted by different organic solvents on growth of human colon adenocarcinoma cells was investigated, and the method of extraction of gliadin with potent inhibitory activity was established.The main results were as follows:1) The gliadin was extracted by80%concentration and pH7of ethanol, ethylene glycol, isopropanol, tert-butanol and acetone, the results showed that extration rate of gliadin extracted by ethylene glycol was the highest of all.2) The gliadin was extracted by50%,60%,70%,80%and90%of ethylene (pH7). the results showed that the extraction rate of gliadin extracted by90%of ethylene glycol was the highest fo all.3) The gliadin was extracted by90%of ethylene at pH6.0,6.5,7.0,7.5and8.0. The results showed that the extraction rate of gliadin extracted at pH7.0was the highest of all.4) Protein precipitation tested at pH3.5,4.0,4.5.5.0and5.5. the highest value at pH4.5.5) The effect of gliadin extracted from apricot kernel by ethanol, ethylene glycol, isopropanol, tert-butanol and acetone on growth of human colon adenocarcinoma cells were investigated respectivly. The results indicated that only gliadin extracted by ethanol showed inhibitory activity significantly.6) The effects of gliadin extracted from apricot kernel through three different methods on growth of human colon adenocarcinoma cells were investigated. The results showed that gliadin extracted by80% ethonal (pH7) with most potent inhibitory activity, and the value of IC50was1.128mg/mL...
