First, Trichoderma reesei Rut-C30cellulase produced by liquid medium andculture conditions were optimized to MA medium-based medium, the use of single-factor test to the basic optimization of media and culture conditionsscreening of themain factors in the medium then use the PB design, the use of climbing test will bethe main factors approaching the optimal value area, and finally the use of centralcomposite design and response surface methodology to determine the optimalmedium, the final fermentation medium (1L) as follows: crystalline cellulose,41.8g19.1g wheat bran, soybean cake powder3.0g, the urea0.38g,(NH4)2SO43.5g KH2PO44.0g, MgSO4·7H2O0.75g, CaCl2· H2O1.2g,20mL of traceelements480mL of pH=5phosphate-citrate buffer, which installed the amount offluid to50mL. Filter paper enzyme activity by model validation has been optimizedfor enzyme production for8.21IU/mL the CMC activity63.64IU/mL, xylanaseactivity27.4IU/mL, glucosidase activity0.96IU/ml; which the filter paper activitythan single-factor optimization the results7.03IU/mL16.78%209.79%than theunoptimized results.Then strain the sodium alginate immobilized into a group of filamentousfungi in liquid fermentation process affect activity to some extent inhibited by themethod of immobilization; and bacteria5L fermenter small scale, small scale enzymeliving is slightly lower than the shake flask level.Finally, the above research results collected to the fermentation broth, and thecentrifugal save as an enzymatic hydrolyzate of corncob biomass hydrolysisconditions, a single factor test, and the optimal conditions for enzymatic hydrolysis ofsugar on the six kinds of biomass curve drawn through the sugar yield curve can beseen in the enzymatic hydrolysis before6h biomass in higher efficiency of enzymatichydrolysis, and then further calculated before6h per gram of biomass per hour, thequality of the production of glucose by comparing the final draw, corn stover afterenzymatic weak base treatment of Trichoderma reesei Rut-C30fermentation brothwith high efficiency.
