Chlorogenic acid is one of the main effective components in Eucommia ulmoides Oliv. It plays many important pharmacological roles especially in anticarcinogenicity and antigenotoxicity. However, the availability of chlorogenic acid is limited because there are many disadvantages in current preparation process of chlorogenic acid with low extraction rate. Thus, the effective extraction and purification of chlorogenic acid from abundant leaves of Eucommia ulmoides Oliv is economic signification.The UV spectrophotometry and thin layer chromatography (TLC) scanning were used to accurately determene content of chlorogenic acid in leaves of Eucommia ulmoides Oliv. Paper chromatography followed UV spectrophotometry is reliable and better stability with lower CV, and simple operation.Main factors influencing the extraction of chlorogenic acid from the leaves of Eucommia ulmoides were studied by single factor and orthogonal experiments with multiple analyses. Optimized conditions of water extracting chlorogenic acid were obtained: the leaves were extracted with water of pH 5.0 at a ratio of 1:12, and taken repeatedly at 70℃for 90 min. The yield of chlorogenic acid was 2.19%.Optimized conditions of ethanol extracting process were as following: the leaves were extracted with 40% ethanol of pH 5.0 at a ratio of 1:16, and taken repeatedly at 60℃for 90 min. The yield of chlorogenic acid was 2.39%. Optimized conditions of ultrasonic wave extracting process were established. The leaves were extracted with 60% ethanol of pH 5 at a ratio of 1:8, and taken repeatedly at the frequency of 20kHz for 40 min. The yield of chlorogenic acid was 2.61%.The yield of chlorogenic acid based on three processes is higher than previously reported in literatures. The processes are promising for chlorogenic acid industrial production and beneficial to the reasonable utilization and further development of Eucommia ulmoides in Shaanxi province.
