Utilizing of large-scale mammalian cell to culture production factors, interferons, h ormones, vaccines, monoclonal antibodies and enzymes with medical value, has becom e the core of the high-tech industrial chain of biopharmaceutical. As the result of the accumulation of metabolic waste, such as CO2, lactic acid, ammonia, urea and indole, the growth of mammalian cells is inhibited and the cost of mammalian cell culture is substantial increased. The key and urgent problem of large-scale biological products is how to timely remove these metabolites, optimize culture environment of mammalian cell, thereby increase the production rate of mammalian cells and reduce the cost of animal cell culture, which has got important application value.In this thesis, the cytotoxicity of nano-attapulgite was studied by means of the exploration of nano-attapulgite’s effect on the Vero (the African green monkey kidney cell) cell morphology, cell viability, cell apoptosis and Lactate Dehydrogenase (LDH). In addition, physical methods were employed to explore the regulated-mechanism of nano-attapulgite with Vero cells Main contents of this work are as follow:(1) Study the preparation methods of nano-attapulgite. Three methods including: sodium hexametaphosphate, hydrochloric acid and sodium polyacrylate were utilized for preparing nano-attapulgite. The TEM was employed to evaluate the methods of nano-attapulgite’s purification and dispersion, then, the zeta potential was measured by the zeta potential tester. The results showed that the nano-attapulgite purified by sodium polyacrylate has the best stability, the second is sodium hexametaphosphate, and hydrochloric acid is the worst.(2) Study the cytotoxicity of nano-attapulgite. With nano-attapulgite, the cell morphology, cell growth rate, cell viability, cell apotosis, concentration of LDH and DNA break were observed and measured, respectively. The results illustrate that nano-attapulgite has no significant toxicity on the mammalian cell. To some degree, nano-attapulgite with a certain concentration can promote the proliferation and growth of the mammalian cell.(3) Desk studies the interaction mechanism of nano-attapulgite with the mammalian cells. FTIR, XPS and BUN kit were employed to study the physical and chemical properties of the Attapulgite fore-and-aft coprocessing with the mammalian cell. The results testified that nano-attapulgite can effectively reduce the concentration of harmful metabolites in the cells, but also optimize the culture environment for mammalian cell, regulate the nutrient’s intake of the mammalian cell, and improve the efficiency of cell growth and proliferation.
