Ferulic acid′s chemical name is4-hydroxyl-3-methoxy cinnamic acid,whichis a generally phenolic acids exists in the plant.Ferulic acid in plants is mainconstitute part of the cell wall with cell-wall polysaccharides and lignincrosslinking,ferulic acid is one of the effective Chinese medicinal cropcomponents such as ligusticum wallichii,angelica sinensis,ferulaasafoetida.Ferulic acid is recognized as a natural antioxidants internationally,andis also regard as cancer awareness material in recent years,with physiologicalfunctions of antioxidation,antiatheroscloresis,antithrombotic and fight bacteriumdiminish inflammation,resistance of mutations,cancer prevention and coronaryheart disease,it is widely used in medicine,cosmetics,health care products andfood additive,etc.Recent studies findings showed that,ferulic acid has a highercontent in rice bran,barley and wheat bran.Wheat bran is the wheat powderresidual part in the process of extraction wheat flour and germ,primarily iscortex,general accounting for about15%of the quality of wheat,in our countrywith an annual production of20million tons,but is mainly used for feed rawmaterials,the economy added value was low.The new study founds,wheat branferulic acid is the highest content of the phenolic acids,it mainly cross-linkingthrough ester bond with the polysaccharide and lignin,either self esterification oretherification form ferulic acid.Extract ferulic acid from wheat bran not onlyincrease the economic value of the wheat bran,but also make the fullexploitation.Therefore,this research take wheat bran as raw materials,discuss differentextraction and purification methods of ferulic acid,and obtained the besttechnology,and identified the purification products.The main research contentsand the conclusions are as follows:a Different ferulic acid extraction methodsThe best extraction technology of ferulic acid were studied respectivelywith alkali liquor extraction,ultrasonic wave assisted extraction, ultrasonic-assisted alkaline-alcohol extraction method.Studied the optimaldodage of the remove part of starch and protein by amylase and protease,in thewheat bran pretreatment,and obtain the optimum condition of alkali liquorextraction,the results are: in order to remove the starch and protein,the dosage ofamylase for240U/g,alkali protease dosage for1200U/g,the best condition for thealkali liquor extraction is: solid-to-liquid ratio for1:15,concentration of alkaliliquor for1%,extraction temperature of80℃,extracting time for2h,under thebest technical conditions the ferulic acid average content is2.47mg/g.The bestextraction for utrasonic-assisted extraction process:extraction temperature of80℃,extracting time for1.5h,concentration of alkali liquor for1%,solid-to-liquidratio for1:12,utrasonic power for225W,ferulic acid average amount of3.13mg/gin the best condition.The best conditions for ultrasonic-assisted alkaline-alcoholextraction:solid-to-liquid ratio for1:12,concentration of alkali liquor for4%,alkaline-alcohol volume ratio of2:1,ultrasonic power225w, extractiontemperature of80℃,extracting time for30min,under this conditions which theaverage ferulic acid extraction for4.27mg/g.b Different ferulic acid purification methodsThe best purification technology of ferulic acid were studie respectively bythe method of macroporous absorption resin and ultrafiltration technology.UsingHPD-100type of macroporous adsorption resin purify wheat bran ferulic acidextraction,determine the optimum technological conditions:adsorption velocityof2BV/h,the maximum quantity of sample solution for seven times the volumeof bed,sample concentration for0.2mg/mL,desorption volume fraction is60%,desorption velocity is1BV/h,the dosage of desorption for six times thevolume of the bed.The optimum technological conditions of ultrafiltrationpurification method:the molecule weight cut-off (MWCO) was10000,ultrafiltration pressure for2bar,extract concentration of0.1mg/mL,ultrafiltrationtime for45-60min.c Identify two kinds of ferulic acid purification product use thin layerchromatography(TLC)and high performance liquid chromatography(HPLC)method,the results showed that two kinds of purification products contain ferulicacid.And macroporous adsorption resin purification of ferulic acid content morethan ultrafiltration method.The ferulic acid content are:91.2%and87.7%, so using macroporous resin adsorption method purify of wheat bran ferulic acidextraction,can get high purity of ferulic acid.
