Mannich reaction is also known as amine methylation reaction, because thesynthetic product obtained Mannich has an extremely important role in the fieldsof medicine, biological, chemical,as Mannich reaction was discovered early byresearchers it had been popular and became an important organic reactions. Inrecent years, with the safety and green of the organic synthesis, the people hadhaven a new understanding on Mannich reaction with a lot of improvement inthe synthesis of materials, catalysts and reaction environment. Initially, materialswas simple aliphatic amines and aldehydes, now people begin to put wide rangeof materials into the Mannich reaction through the exploration and research inthe field of chemistry, which also greatly improve the pharmaceuticalantibacterial activity of the products. There are many Catalysts, such as theLewis acid, Lewis base, and they can effectively accelerate the reaction rate andimprove the yield.Ferrocene derivatives has been researched extensively as manyphysiological activities,2-amino-5-disubstituted-1,3,4-thiadiazole is a typicalrepresentative in heterocyclic amines, It is an important intermediate of themedicine and pesticide, it has antibacterial, bactericidal, anti-inflammatory,antitumor and other biological activities. In this experiment, both of them wereintroduced into the Mannich reaction to synthesis the Mannich base containingferrocenyl and heterocycles, and the antibacterial activity of the product wastested. Experiment was divided into four parts as following.(1) Refer to the corresponding literature, Ferrocene, phosphoric acid andacetic anhydride were taken as material, and synthetize the acetyl ferrocene.(2) Synthesis of the2-Amino-5-substituted-1,3,4-thiadiazole were taken.When Synthetize2-amino-5-aryl-1,3,4-thiadiazole,we took the phosphorusoxychloride as catalyst and the production rate was high. (3) Taking into account the economic question and other reasons, the Lewisacid-sulfuric acid as catalyst were chosen when synthesis of the final productwas begun. Formaldehyde, acetyl ferrocene and2-amino-5-substituted-1,3,4-thiadiazole were the materials,11kinds of Mannich base were synthetizedthrougt one-pot method. All compounds were characterized by melting point, IR,1H-NMR and13C-NMR to determine their structure.(4) Antibacterial experiments was tested by the filter paper method,Escherichia coli, Staphylococcus aureus, Saccharomyces cerevisiae,Streptococcus, Actinomycete were choosed for the test. Experimental datasshowed that the all products had the antibacterial activity. In5-substitutedcompounds, the antibacterial activity of the aryl compounds was significantlybetter than the aliphatic compounds...
