Microbes are the most common biological beings in the living environmentand also are the creatures which contact with the human beings most. Microbialpollution could influence physical health, even may cause lesions if the effect getworse. Because of the special physiological characteristics of human feet, shoesbecome the most serious parts of clothes in the microbial pollution. With theprogress of science and technology and the improvement of the life quality,antibacterial industry gradually developed, it has decades of history yet. But thefast development also causes some problems: the antibacterial shoe materialmarket is in disorder, the classes, quality of antibacterial products are different,and their authenticities are difficult to distinguish; some manufacturersexaggerate propaganda, mislead the consumer; some manufacturers even ignorethe environment pollution and human safety, add excess agent to aspire the highantibacterial effect. Antibacterial detection methods were mature and numerous,the antimicrobial effect of Different materials need to use different methods fortesting, but these different methods for the same material are not comparable. Atpresent, test methods of antibacterial properties for shoe materials are related toonly a few standards, and it has not been specifically researched. In view of thissituation, this paper aims to explore and establish a unified antibacterial testmethod and mould proof s for shoes materials.Generally, the paper has three parts: the selection, improvement andevaluation for the methods.First of all, select suitable antibacterial or mould proof detection methods forshoe materials. There are6kinds of footwear materials, textiles, EVA, PUmidsole, PU synthetic leather, leather materials, HPVC. According to thestandard method, absorption method is finally chosen to be the textile antibacterial detection method. Confirmed by the experiment, other shoematerials should adopt the first one between flask shaking method and the filmmethod. Because of the material properties, there will be difference in the resultsof antibacterial detection by sticking film method. By comparison with othermaterials (paper, feed, plastic) and the US anti-mildew performance test, moldperformance detection method of shoe materials was finally chosen. The methodis: inoculation fluids dropping on the samples and incubate it in Petri dishes instatic culture.In order to make the experimental methods and results more comparable,standard samples of each kind of material which are divided into differentantibacterial property levels were respectively prepared by experiments, and foreach concentration of antimicrobial samples, there were antibacterial propertiesand mildew-proof performance test. And the results are analyzed by one wayANOVA to evaluate the homogeneity of the samples; it shows that the uniformityof antimicrobial properties of samples is qualified.This was followed by the improving of detection method. By analyzing themethods that chosen for shoe materials, it has been found that conventionalmethods are time consuming, disaccord of operation process parameters, and therepresentation is not unified and scientifical. Since the traditional methods haveall these defects, improvements are researched by experiments.(1)First toimprove the timeliness, the previous two or three steps of bacterial cell brothpreparation pre training program were turned into a one-step bacterial brothshock, two-step fungal broth shock procedure for incubate, greatly shorten theexperiment time. And innovative use the mixed bacteria liquid, simplifiesexperimental steps greatly, it is more consistent with the actual life situation.(2)Secondly introduce ATP rapid fluorescence detection method instead of platecount, and propose the use of a simple shoe sterilization method. Theexperimental time can be reduced from the original4days to2days. In theprocess of detection using absorption and oscillation method, several importantfactors were optimized: sample specifications, training time, added liquidvolume, elution volume. The experimental results are analyzed and scientificallycompared, depend the situation, choose the relatively rational and effectiveresults representation method. Finally we evaluated the improved antibacterial detection methods,determined the stability of strain, culture medium and solution used in theexperimental process. In a month of the warranty period, compared withconventional methods, the improved training program for liquid bacteria do nothave too big change in the way of bacteria growth. The only difference is, tomeet the concentration, the improved bacteria levitation liquid the cultivation oftraining program with time strains of the storage time increased, but does notaffect the whole time. Repeatability of determination result represented by thebacteria value is good, and the conclusions can meet the requirements ofantibacterial performance presentation.
