Layered Double Hydroxides (LDHs) are one type of biocompatibleinorganic multifunctional materials, which is often used as powdermaterials to carry the drug molecules, biological molecules DNA orsiRNA and release slowly or selectively in vivo. In addition, Hydroxy inthe surface of LDHs can be immobilized protein without modificationand maintain the protein activity supplementing the shortcomings of theorganic substrate. LDHs microarray film can be obtained easily andimmobilized cells selectively which is a potential support material withgood biocompatibility for selective cell adhesion, cell migration and celltransfer et al. In theory, LDHs microarray film can be used as a simplesubstrate materials of the Protein Microarray Technology. In this presentstudy, LDHs patterned arrays of different sizes was fabricated in modified copper, gold, glass substrates.The pH biosensor and the adhesion andselective growth of Hep G2cell were achieved. The details involved arepresented as follows.1. LDHs film in copper and gold substrates (LDHs/Cu, LDHs/Au)was fabricated by hydrothermal method and the morphology, growthmechanism and hydrophobic properties were investigated. Thefluorescent protein (BSA-FITC) was immobilized on it selectively whichdetects pH changes in environment. The results showed that LDHs/Cufilm can be used to adsorb fluorescent protein molecules BSA-FITCwithout modification (BSA-FITC/MgAl-LDH/Cu). The fluorescenceintensity of BSA-FITC/MgAl-LDH/Cu and pH is a linear relationshipbetween pH9-10.5which is a potential two-dimensional biosensor in thecellular microenvironment.The small size of the LDHs microarray and anti-microarray film inthe gold substrate (LDHs/Au) was fabricated by hydrothermal methodand Lithography overcoming the shortcomings of copper dissolved andentered the LDHs layers. The fluorescent protein (BSA-FITC) was alsoimmobilized on it selectively. The results showed thatBSA-FITC/MgAl-LDH/Au is remarkably sensitive to the variation of thepH in environment and also capture the minute changing between pH9.5-10.2. The LDHs microarray film in the glass substrate modified by DMABS (LDHs/DMABS/glass) was fabricated by hydrothermal methodand Lithography. The growth of Hep G2cell were investigated. Theresults showed that LDHs film can adhere Hep G2cell and cannot inhibitthe cell activity. The selective growth of Hep G2cells was achieved bydifferent distribution of DMABS and LDHs film in the glass substrate.
