Objective: The aim of the present study was to assess the predictive value of theMN test, comet assay and the activity of SOD and GSH-Px in Carassius auratus redvariety as biomarkers for in situ monitoring of phenol pollution.Methods: Carassius auratus red variety was exposed to phenol at differentLevels (0,2.16,4.31,8.62,17.24mg/L). MN frequency and trailing rate in erythrocytes,the activity of SOD and GSH-Px in gonad were measured in exposure time of24,48,72,96hour.Results:1. LC50of phenol to Carassius auratus red variety at24,48,96h was38.02,36.83和34.48mg/L, Sc of phenol to Carassius auratus red variety was3.45mg/L.2. Carassius auratus red variety were exposed for96hours to0,2.16,4.31,8.62,17.24mg/L of phenol. MN frequency was determined in fish erythrocytes. Significantincrease in MN frequency was observed in erythrocytes of fish exposed to phenolwith the exposure time lasting, with the concentration of phenol increased, the rate ofmicronucleus increased and then decreased, The study revealed that micronucleus test,as an index of phenol exposure, appears to be a sensitive biomarker to evaluate phenolpollution in Carassius auratus red variety under controlled conditions.3. The assay was performed on whole blood at24h,48h,72h and96h exposure intervals. A significant increase in DNA damage was observed between the treatment andcontrols. However, the percentage of DNA damage decreased at72h exposurecompared with48h exposure and decreased at96h exposure compared with72hexposure.4. The effects of phenol to the activity of SOD and GSH-Px of Carassius auratus redvariety in different levels for24h,48h,72h and96h: The SOD activity was increasedthen decreased, was induced to the maximum earlier with the concentration of phenol increased; with the exposure time lasting and concentration of phenol increased, theactivity of GSH-Px had not significant difference compared to the control group.Conclusion:1. LC50of phenol to Carassius auratus red variety in24,48,96h was38.02,36.83和34.48mg/L, Sc of phenol to Carassius auratus red variety was3.45mg/L.2. Except for the activity of GSH-Px in gonad, MN frequency and trailing rate, theactivity of SOD in Carassius auratus red variety are suitable as biomarkers of phenolpollution.3. Carassius auratus red variety could prove to be interesting models for investigationsof phenol pollution because of their sensitivity.
