In recent years, fluorescent probe has become the focus of research topic, it hashigh selectivity, sensitivity, optical fiber remote detection as well as the naked eyeidentification and other advantages, so his increasingly extensive application.Indolium tricarbocyanine dye is a kind of near-infrared(NIR) dye, can avoid thematerial self absorption and fluorescence background interference, the fluorescentprobe has great application prospects. Similarly the prodrug is also the focus ofattention, the prodrug can overcome shortcomings and can increase drug drug efficacy.The research work of this thesis is mainly based on the dyes and prodrugs, is dividedinto three chapters. Main class capacity is as follows:In Chapter I, We hanve first introduced the basic concept, fluorescence influencefactors and the study on the principle of fluorescence of PET theory, then introducesthe definition, classification, prodrugs advantages, porphyrin anticancer prodrugmechanism and research progress and photolysis of protecting groups and researchdevelopment of porphyrin synthesis method, finally puts forward the the design ideaand research contents.In Chapter II, we based on the fluorescence of PET principle design andsynthesis of a near-infrared Cu~+fluorescent probe. When Cu~+is added to the probe inthe liquor, probe and indolium tricarbocyanine dyes maternally linked to the nitrogenatom and Cu~+binding, probe the ultraviolet absorption peak red shift. With Cu~+entry,probe photoelectron induced transfer effect is suppressed, the fluorescent probegradually strengthened and when the probe/Cu~+concentration ratio of1:1when thefluorescence increases to a maximum. The probe of Cu~+with high selectivity andsensitivity, not by other ion effect. Probe to the low detection limit, can in thephysiological range of pH very good applications, are also believed to be in thebiology application. So far, for near-infrared Cu~+fluorescent probes reported verylittle, we therefore probe photoelectron induced transfer effect to design Cu~+is verymeaningful and valuable.In Chapter III, synthesis and characterization of optically controlled release ofporphyrin anticancer prodrug made17and22as well as a series of intermediatecompounds,17and22are porphyrin derivatives and contains the o-nitrobenzyl,wherein the porphyrin plays on cancer cell positioning, the o-nitrobenzyl is a photolysis protection base, at certain wavelengths of light irradiation, will release theprotected tc.17and22of its water solubility is very poor, limit its application, needto be further improved.
