Sulfhydryl substances widely present in vivo,and play heavy metal detoxification, antagonistic to ionizing radiation, remove free radical; and body growth and development,slow down aging and certain diseases. Therefore, the establishment of simple,fast and convenient determination of the thiol substance is of great significance.This article from the following three chapters are discussed in detail.Chapter1IntroductionSummarized in a form of existence and thiol compounds in vivo and its physiological role, and pointed out the importance of thiol compounds, and summarizes other scholars determination of thiol compounds, and put forward ideas and research of the topics.Chapter2Determination of edible mushroom fungi in the thiol content of substancesScreening of crops rich in thiol peptide as exogenous supplements to prevent and cure diseases more economical, more effective than the use of chemically synthesized preparations. In this study, the content of the market in the common edible mushroom thiol substances to make a detection. At present, the content of thiol compounds in biological samples determination mostly involved in the process of cell broken, solvent extraction, etc. before processing, during the in evitable by the oxidative chemical environment of, Reduced Thiol by chemical modification and degeneration, lead to determination results can not response to biological the sample itself, containing thiol compounds was content and even lead to missed. This study is based on thiol derivatization reagent as a probe, proposed a derivatization reagent and mushroom samples synchronized ultrasonic broken extraction of thiol compounds before the new sample processing method, and compared with the general sample preparation method. The experimental results show that:The new law can be more complete and comprehensive characterization of the organism’s cells, thiol substances. And o-phthalaldehyde (OPA) derivatization reagent glutathione (GSH) conducted a qualitative, quantitative analysis. This study is to develop a simple reliable biological samples containing thiol peptides in situ analysis methods for a new attempt and explore.Chapter3Electrochemical Properties of5,5-dithiobis(2-nitrobenzoic acid) and Application to the Determination of GlutathioneElectrochemical properties of5,5-dithiobis(2-nitrobenzoic acid)(DTNB) are investigated using glassy carbon electrode with cyclic voltammetry. Sensitive reductive current is observed by reduction of disufulde bond on the electrode. Influences of acidity, initial potential, reduced glutathioe (GSH),cycling number on the cyclic voltammograms of DTNB have also been investigated and electrochemical mechanism of DTNB on the glassy carbon electrode have been proposed accordingly. Linearly decreasing reduction current of disulfide bond of DTNB with the increasing concentration of GSH have been observed. According to the linearship between reduction current and GSH concentration, novel electrochemical detection method of GSH are established. The determination results are consistent with those determined by spectrometric method.
