The word of proteome originated from the combination of two words of protein and genome, which means a set of protein used to express a kind of gene. The study of proteome not only offers the material basis for the law of lifecycle, but also offers the theoretical basis and resolvent for the interpretation of many morbid mechanisms. Moreover, proteome and nucleic acid are the molecular targets of design of new type of medicine. Because it is helpful to seek the targets of medicine, design the new type of medicine and study the healing priciple and toxicity of medicine, etc., it is of special value to study the interaction of medicine and biomolecule. At present, the environmental pollution, especially the food pollution, is getting more and more harmful to the health of humanity. The determination of food is one of important tasks, which can regulate market economy order, ensure the natural function of economy and insure the physical health and life security of people. Therefore, a complete of determination methods, which have high precision and low detection limit, are needed urgently to prevent the harmful food from entering the market.hi this paper, according to above two objects, using molecule spectroscopy (fluorescence and Uv-Vis spectroscopy) and ion chromatography method, we study the interaction between drug and biomolecule, and the determination of harmful substances in food, respectively. There are three chapters.First, we studied the interaction of Human Serum Albumin, Bovine Serum Albumin and drugs such as epirubicin, daunorubicin and terazosin. Applying fluorescence quenching method and energy transfer theory, we have studied the quenching mechanism of protein and drugs. The binding constants were obtained in the presence and absence of metal ions. According to the thermodynamic parameters, the main sorts of binding force can be known. In addition, using the synchronous fluorescence technology to reduce the interpretation of serum albumin, we successfully determined the concentration of drug, with the high sharpnessof separation and low detection limit.Second, there is energy transfer between ciprofloxacin and Tb3+ and the complex can emit the characteristic fluorescence of Tb3+, so the complex of them can be used to determine DNA as a kind of fluorescent probe. In addition, the hydrophobic environment can dramatically increase the fluorescent intensity of complex. In this part, we studied the fluorescence increased effct of nucleic acid on the complex system, explained the interaction mechanism, put forward the model of interaction between nucleic acid and the complex, and set a method used for selective determination of DNA.Third, sodium sulfoxylate formaldehyde is a kind of bleaching powder used in industry, which is prohibited from adding to food. In this part, we take it as an object to study, by using ion chromatography instrument which is equipped with autech DS-PLUS?auto suppressor and Allsep A-2 anion column as well as conductivity detector. Because some mixtures are difficult to filter, we explored two different methods used to do with the different samples. In addition, we also explore the opitimum chromatography conditions and the basis in the qualitation analyst and quantition determination.
