Rhizoma Dioscoreae from Edong is the tuber of Dioscoreaceae Plant Dioscorea japonica Thund. There are rich resources as industrial crop in Edong Hubei province. The literature recorded that the tuber of the Dioscorea japonica Thund is not only can edible but also can serve as promoting digestion medicine. However, the Rhizoma Dioscoreae from Edong is only served as vegetables that the medicine value is remain to be developed. The quality analysis、 pharmacology and extraction technology for polysaccharides of Rhizoma Dioscoreae from Edong have not been reported. We carry on comprehensive study for the Rhizoma Dioscoreae from Edong, and provide certain theoretical basis for medicinal use of Rhizoma Dioscoreae from Edong. At the same time, the study aim is establishing the basis for utilization and exploitation of plant resources.The character、microscopic identification、Thin-Layer chroma-tography、test、extract and the determination of content were used in quality analysis. We determined the contents of polyphenolics、 polysaccharides and total saponins by UV-VIS spectrophotometry. At the same time, we determined the contents of allantoin by HPLC, and the content of allantoin in Rhizoma Dioscoreae from Edong is higher than that in Rhizoma Dioscoreae. The accommodation of immune activities with polysaccharedes was carried on. The results are as follows:at the dose of100mg·kg-1, the effects of polysaccharides towards normal mice on the function of macrophage cell in abdominal cavity were not obviously promoted, the index number of the immune organs had an increase at a certain extent. At the dose of200mg·kg-1, polysaccharides can partly increase the phagocytic index and phagocytic percentage in peritoneal macrophages and significantly increase immune organs index in immunosuppressed mice. At the dose of400mgkg-1, polysaccharides can significantly enhance the phagocytosis in peritoneal macrophages and the immune organs index in immuno-suppressed mice.To study on the antioxidant activity of polysaccharides in Rhizoma Dioscoreae from Edong by using the Fenton system. The conclusion was:it had good ability to scavenge hydroxyl radicals, and the scavenging ability related with the concentration of the polysaccharides at a positive correlation. The IC50is1.96mg·ml-1.This paper mainly includes the studies on conditions of extraction and purification for polysaccharides from Rhizoma Dioscoreae from Edong. The extraction yield was selected as the inspection index. We optimized the extraction technology by orthogonal test on the basis of single factor experiment. The optimum conditions for the extraction of polysaccharides are as follows:temperature100℃, solid-liquid ratio1:10, time120min and ethanol concentration80%. Under the conditions, the extraction yield of polysaccharides is58.32mg·g-1, the content of polysa-ccharides in crude polysaccharides is41.05%.
