Polygala japonica Houtt has the effect of expectorant cough, blood circulation, detoxification, swelling and so on. With broad application prospects in the field of drug development.There are frequent reports on the research of Polygala japonica Houtt, the domestic reports mainly concentrated in the compound preparation and clinical application studies,while the foreign reports mainly concentrated on the extraction and separation of effective components, structure identification of Polygala japonica Houtt. However, there are no reports on the research of the Liquor of Polygala japonica Houtt through biological transformation at home and abroad.To take full advantage of natural plant of Polygala japonica Houtt, and to make research and development of its new formulation, the paper tries to extract fermented pharmacological activity of total saponins from Polygala japonica Houtt by using traditional fermentation methods to make oral liquor.We also made a systematic investigation of the preparation technology, quality standards, the initial stability and pharmacological activity.The main content of this study and its conclusions:In preparation, We make use of traditional methods to fermentate, extract oral liquid from Pharmacological activity Polygala japonica Houtt , optimize the preparation process of oral liquor by orthogonal tests, as a result, the preparation process by orthogonal test was proved the best. Best conditions: Add water, 8 times of the total saponins, add sugar,1 times of the total saponins, pH7.0, let stand for 10minutes,repeated the experiments for many times by the selected process, as it's proved that the experimental method is simple, and the oral liquor is stability, and meets the quality standards of oral liquid preparation.On the research of quality standards, we made a chromatography of the Polygala japonica Houtt and the oral liquor respectively, the experimental results proved that the selected method is specific,Qualitative methods can be used as identification.On this basis, we made a stability study of the oral liquor, and carried out an experiment at room temperature, accelerated test (40℃, relative humidity 75%), and other factors in the study trial, the indicators didn't change significantly, indicating a good stability.Negative control without interference; prove that the selected method is specific. Qualitative methods can be used as identification.In pharmacological research, we studied on the in vitro antibacterial activity,pharmacology and toxicology, analgesic effect and anti-inflammatory activity:We compared the antimicrobial activity of the oral liquor through the filter paper with different concentrations, the oral liquor has proved to have some inhibitory effect, as the oral liquor get concentrated, the inhibitory effect enhanced gradually.Among them, the inhibition of Staphylococcus aureus is most obvious.Agar plates by serial dilution method determined the minimum inhibitory concentration (MIC), melon seeds fermented oral liquid gold Staphylococcus aureus, Escherichia coli, Candida albicans, Pseudomonas and Bacillus subtilis were the minimum inhibitory concentration 25mg/ml, 50mg/ml, 50mg/ml, 50mg/ml and 100mg/ml; liquoer oral, Staphylococcus aureus fermentation minimum inhibitory concentration of the smallest, with a significant antibacterial activity.Oral liquid on the acute toxicity test in mice showed that oral liquid drug is toxicity, safety range, for clinical use.Pain caused by hot plate method and acetic acid reaction of writhing induced analgesic effect of oral liquid on the study results showed that each dose oral liquid all have analgesic effects, as the concentration increases, the analgesic effect is more apparent.Xylene by the classical ear swelling test in mice for anti-inflammatory effect of oral liquid experimental,the results show that the gold seeds are fermented oral dose of the mouse ear swelling has obvious anti-inflammatory effect, but with the oral liquid concentration increases, the more obvious anti-inflammatory effect.In summary, oral liquid fermented seeds of gold, the traditional fermented by pure extract of Chinese medicine made of the effective area, safe, effective, long-term use, but also for a wide range of people, and the use of modern scientific methods to control the preparation of quality plays a social and economic benefits for future development of a drug side effects and convenient application of pure Chinese medicine.
