Corn gluten meal(CGM) is a byproduct with the highest protein content in corn wetmilling.Zein is the main protein in CGM.Because of its poor solubility and imbance in aminoacid composition,the application of CGM was restricted in food industry.Zein contains highproportion of leucine,isoleucine,alanine and proline,which provides a good advantage forpreparation of peptides with bioactivity.Anti-alcohol products with high contents of leucineand alanine are manufactured by using specific enzymes and conditions of enzymatichydrolysis combine with modern bioseparation technology,which can find new ways toincrease use and economic value,make corn protein resources to get more reasonableuse,reduce environmental pollution,as well as raise economic benefits of corn processingenterprise.CGM was used as raw material and was modified by enzymes in thisdissertation.Decolouring technology of hudrolysate from CGM with activated carbon wasoptimized.Then peptide drinks with bright colour,uniformity,good flavor were obtained fromhydrolysate of CGM and corn peptide powder,respectively.Portable tablets were producedthrough wet granulation.Finally,the animal efficacy experiment of corn peptide was carriedout.The results were as follows:Decolouring technology of hydrolysate from CGM with activated carbon and itsmolecular weight distribution were investigated.Decolouring conditions of hydrolysate wereoptimized by response sruface analysis on the base of single factor experiment,and molecularweight distribution of corn peptides before and after decolouring were compared by molecularexclusion chromatography.The results showed that the optimum decolouring technology werepH of3.13,activated carbon concentration of1.25%,temperature of45℃,reaction time of1h.Under these conditions,decolouring ratio and protein recovery yield reached86.24%and88.96%,respectively.Molecular weight of corn peptides in the hydrolysate mainly distributedbetween4677.89Da~246.59Da before decolouring,while between2976.23Da~250.80Daafter decolouring.By means of sensory quality and centrifugal sedimentation rate,the suitable formula ofpeptide drink from corn peptide powder was determined by single factor experiments asfollows:peptide powder2.5%,soft sugar5.0%,citric acid0.6%,sodium citrate0.5%,sodiumcyclamate0.1%,CMC0.09%,-cyclodextrine0.3%. Sensory quality as marking criterion,the optimum formula of peptide drink fromhydrolysate of CGM was confirmed by orthogonal experiment as follows: hydrolysate ofCGM94.587%,sucrose4.0%,citric acid0.5%,CMC0.2%,sodium citrate0.5%,sodiumcyclamate0.2%,apple essence0.013%.The final formula of tablets was obtained by using single experiment to investigateeffects of each factor on preparation through wet granulation.And final formula was cornpeptide powder62.19%,starch18.75%,dextrin18.75%,95%ethanol25%(w/v),magnesiumstearate0.31%.Preparation process as follows:corn peptide powder,starch and dextrin weremixed together,then soft material was prepared by adding and homogenizing95%ethanolquickly,particles,which obtained through40mesh sieve in advance,were reprepared through40mesh sieve after drying20min at60℃,tablets were produced after addition of magnesiumstearate finally.The results of animal efficacy experiment of corn peptide showed that,compared to thecontrol group, blood ethanol levels in corn peptide groups(not ultrafiltration and ulfiltration)were decreased by60.85%and63.61%respectively after intragastrical administration1h,which showed strong facilitation on alcohol metabolism in mice.Free glutamic acid andbranched chain amino acids in plasma,especially leucine might has synergistic effect onalcohol metabolism in mice.
