In the present study the walnuts as the main raw material, the first in mechanical broken based on the three enzymes of the complex legal soxhlet to oil on walnut is to determine enzyme solution to peptide content and enzyme solution to the liquid DPPH clearance for index, the enzyme solution were the best complex protease; The complex protease adding amount of crude protein concentration, walnut, enzymatic hydrolysis temperature, enzyme solution liquid pH and enzyme solution time on enzyme-hydrolysed effect the influence, and using the orthogonal experiment, optimize the process conditions; Then will the walnut meal product enzyme solution through Sephadex G-25gel chromatography column for separation, and the molecular weight distribution and each separate components of O2-· clearance,·OH clearance, DPPH· clearance and total antioxidant capacity research on strong antioxidant activity of the components of the separation and enrichment of freeze-drying YingYangXing made of walnut peptide powder.The results show that:Neutral protease, alkali protease, enzymatic according to1:2:3blends with walnut meal for enzyme solution when the best composite enzyme, best enzyme solution conditions for:the enzyme concentration for5000U/g, substrate concentration is3%, and the temperature of65℃, pH7.2, enzyme solution time for5h. In the condition of experiment, walnut meal composite enzyme solution of liquid concentration of peptide1.30mg/ml, DPPH clearance for93%.Sephadex G-25gel chromatography column of separating out with strong oxidation resistance of peptide fragments of molecular weight mainly focus on5733-307-u between.
