Xanthohumol is a prenylflavoniod of simple structure from Humulus lupulus L, it isonly in the hops and it is the main flavonids. The main effective components for brewing beer are hop resin and hop oil. The technology of supercritical CO2extraction can realize the extraction of effective comonents and the exteacts is stable、easy to store and have a higher utilization rate, so the rate of hop exteacts was increased in the beer production year by year. But xanthohumol can’t extract by supercritical CO2extraction and remain in the supercritical CO2extration residue, these residue as a waste and can’t available use. Therefore, this article mainly study extraction, purification of xanthohumol from supercritical CO2extration residue of Humulus lupulus and its effects of anti-oxidation and hypolipidemic through mice experiment. It was studied for the future medicine of preventing atherosclerosis and providing a theoretical basis for its development and reuse of waste hops.The main content and the results are as follows:(1) This experiment studied the extraction of xanthohumol through ultrasonic extraction and constant gas bath concussion extraction, the experiment results show that ultrasonic extraction is better than constant gas bath concussion extraction.(2) This experiment studied the extraction of xanthohumol by methanol, ethanol and acetone as extraction solvent from waste hops, extract was detected by HPLC, the results show that the content of xanthohumol was the highest through acetone as extraction solvent, the content was quite through methanol and ethanol, but considering the problem of solvent residual, the experimental choose ethanol as extraction solvent.(3) This test validated several different ways of separation and purification of xanthohumol, including macroporous adsorption resin method, organic solvent extraction method, diatomite adsorption method, sodium carbonate salting method. The highest of xanthohumol purification was organic solvent method (51.2%), but low yield (19.34%), the highest yield was macroporous adsorption resin method (93.56%), but low purity (11.6%). So in later experiment, first preliminary separation XN using macroporous adsorption resin method, then further purification using organic solvent method.(4) Through the hyperlipidemia mice test in vivo, XN low doses (2mg/10g bw· d)、middle doses (6mg/10g bw· d), high doses (12mg/10g bw· d) could significantly reduce TG of serum and could significantly rise HDL-c of serum,showed that XN reduced hyperlipidemia significantly; when hyperlipidemia mice foraged XN, plasma MDA reduced significantly, liver SOD increased, plasma and liver CAT increased, which showed xanthohumol had antioxidant effect; high doses (12mg/10g bw· d) could lead to cholesterol metabolism disorders; when the normal mice foraged middle dose (6mg/10g bw· d) XN, lipid metabolism in vivo had no influence; when the normal mice foraged middle dose (6mg/10g bw· d) XN, plasma GSH-Px, CAT significantly reduced, liver MDA increased significantly, SOD、GSH-Px significantly reduced, which showed that XN had oxidative damage effect to mice.(5) The paper studied the Dallas mavericks thymus DNA damage of different concentration xanthohumol through the spectrophotometry and electrophoresis, the spectrophotometry showed that when XN concentration in a certain range (concentration of XN less than0.5mg/ml), XN had inhibition effect to DNA damage; the electrophoresis result showed that when XN concentration between0.1-1.5mg/ml, XN had inhibition effect to DNA damage, and with the increase of XN concentration, the inhibition effect to DNA damage gradually strengthened.
