Ginger (Zingiber officinale Rose.) is a widely used medicine and food dual purpose plant which has many pharmacological activities. For example, it can be used to loose cold, stop vomiting, resolve phlegm and relieve cough. Because it has various kinds, a wide range of sources and nutrition and health function, ginger has a very important value for development and broad application prospects and has caused wide attention both at home and abroad. There is an old saying that "the older it is, the hotter it is". This paper introduced a systemic study on analysis of chemical components in seed ginger and tender ginger and antioxidant activity of seed Ginger, based on the Laiwu ginger material, to provide methodological and technical supporting for its development and utilization(1) Analysis of chemical components in seed ginger and tender gingerThis experiment did a systemic analysis of volatile components, gingerol-related compounds and nutrient composition in seed ginger and tender ginger. Firstly, volatile components from seed ginger and tender ginger were analyzed by headspace solid phase microextraction (SPME) combined with GC-MS. Secondly, the gingerol-related compounds in seed ginger and tender ginger were analyzed by high performance liquid chromatography. Comparing the volatile constituents of seed ginger and tender ginger, it showed that the main composition and components were similar. From the results, we also can know that the contents of6-gingerol,8-gingerol and10-gingerol in seed ginger are two times, one and a half times and two times those in tender ginger separately. The water capacity of seed ginger and tender ginger is very similar, however, the content of amino acid and vitamins in seed ginger are both lower than those in tender ginger and the content of crude fiber in seed ginger is higher than that in tender ginger. From this study we can know that seed ginger has more obvious medicinal value than tender ginger, so it can be used for the extraction and preparation of gingerol-related compounds or in the field of new medicine research and development. However the edibility of tender ginger is higher than seed ginger, so tender ginger can be used in food products which take ginger as the main raw materials.(2) Analysis of gingerol-related compounds in seed gingerThe objective of this study is to establish a rapid method for the analysis of gingerol-related compounds in seed ginger by using high performance liquid chromatography coupled with electron spray ionization-quadrupole-time of flight mass/mass spectrometry (HPLC-ESI-Q-TOF-MS/MS). The gingerol-related compounds in seed ginger was separated by an Inertsil ODS-SP column (250mm×4.6mm,5μm) using a binary eluent under gradient conditions. The analytes were detected by ESI-Q-TOF-MS/MS in positive ion mode to obtain MS and MS/MS spectra together with extracted molecular weights. From the MS data, the accurate molecular weights of gingerol-related compounds were obtained; and from the MS/MS data, the (+) ESI-Q-TOF-MS/MS fragmentations were obtained. Results:25gingerol-related compounds were identified from seed ginger(Zingiber officinale Rose.) by attentive studying on the mass spectra of compounds and comparing with reference data that had been reported in some related papers; and the method was certified to be accurate and reliable by comparing with the reference compounds. According to this study, a rapid method for the analysis of gingerol-related compounds in seed ginger (Zingiber officinale Rose.) by using HPLC-ESI-Q-TOF-MS/MS was successfully established. Because of the advantages of no reference substance, rapid measurement and accurate determination, the method of HPLC-ESI-Q-TOF-MS/MS can be used for the rapid analysis of gingerol-related compounds in seed ginger (Zingiber officinale Rose.) and provide technical support for quality control of seed ginger.(3) Studies on the isolation and evaluation of anti-oxidation chemical compositions in seed ginger (Zingiber officinale Rose.)The objective of this study is to isolate and evaluate the anti-oxidation chemical compositions in the methanol extract from seed ginger (Zingiber officinale Rosc.). Compared with ascorbic acid, antioxidant activity of the nine extract groups which were separated from the methanol extract of seed ginger by silica gel column chromatography had been measured and evaluated by four in vitro models, including DPPH radical scavenging, reducing power, hydroxyl radical scavenging and superoxide anion radical scavenging. The content of flavonoids and total phcnolic were also determined. The results indicated that the nine extract groups were greatly different in the antioxidant activity showing a positive correlation with the content of active components. From this study we can know that seed ginger has great antioxidant activity and silica gel column chromatography is a usefull method to isolate anti-oxidation chemical compositions from seed ginger.
