The cottonseed protein and chicken protein is rich in amino acids, traceelements,constant elements, vitamins and some unknown growth factors, then theyare very cheap, but the utilization rate of animal digesting absorption is very low. So,they should actively research a simple, feasible, economic and effective method ofhydrolysis, and change the large amounts to digestible proteins and complex aminoacids. They will be applied directly as animal feed additives,or will be easy to getcheap raw materials of purified amino acids. Thus if treated properly they will greatlynot only improve the utilization and the use value of wastes, but also protect theenvironment.In this papers, because the protein is content of constant nitrogen,they measurethe total nitrogen in cottonseed protein and chicken protein by kieldahl method,andcalculated the protein content. The protein colors with Coomassie brilliant blue.Theabsorbance in the cottonseed and chicken protein fermentation is detected by UVspectrophotometry.And they can calculated the protein concentration by therelationship between standard and protein concentration.They adopt the method ofenzyme and microbial fermentation hydrolysis to degradate cottonseed protein andchicken protein,then they observe the influence of substitutability (%), pH, reactiontemperature (℃) and reaction time (h) to the hydrolysis of neutral protease AS1.398and papain papaya.And they design orthogonal combination to optimize hydrolysisprocess, some conclusions are as follows:1.They measure by kieldahl method that: the protein content in cottonseedprotein is45.4%, thus it is65.8%in chicken protein.2. When the protein concentration in the fermentation varying from0to16.67μg/mL, the concentration have the preferred line relation with absorbance. Thestandard equation of bight Y=0.044x+0.09is needed in the measure. The correlationcoefficient R=0.999, the RSD was3.86%(n=6), the relative standard and deviationis less than4%. 3.Through the hydrolysis process of neutral protease AS1.398and papainpapaya to the cottonseed and chicken protein, we find that the effection of is betterthan neutral protease AS1.398, and the hydrolysis rate of soluble protein reach highly36%. So, as for the papain papaya, its suitable condition is pH=8.0, reactiontemperature for70℃, reaction time for8h and enzyme quantity for2.0%.But as forthe neutral protease AS1.398,it is better to chicken protein, and the hydrolysis rate ofsoluble protein reach highly37%, then the best hydrolysis conditions is pH=8.0,reaction temperature for70℃, reaction time for8h and enzyme quantity for2.5%.4. By means of microbial fermentation of cottonseed protein and feather proteindegradation, and the enzymatic method, you will find that the degradation of themicrobial fermentation is better than the enzymatic method, and the operation is easier,they can guarantee that the quality of the product is not only damaged, but also it canimprove the rate of hydrolysis of soluble proteins, which reaches pectively43%and41%.
