Soybean is the most nutritious kind in beans. It is in the top in hundreds of naturalfoods, for it’s rich in unsaturated fatty acid, microelement, vitamin, high qualityprotein and so on. Soybean is one of the abundant resources in our county. And thehigh demand of soy isolated protein brings a mass of whey wastewater. At the presenttechnology condition in our country, every1t soy isolated protein produced willproduce60~80m3whey wastewater. In recent years, antinutritional factors especiallysoybean trypsin inhibitor (SBTI) attracts attention for its various physiologicalfunction. In addition, SBTI is effective to reduce weight.With the improvement of living, people’s diet structure and habits have changedgreatly, besides the decrease of exercise, obesity increasingly puzzled people. WHOdefinitely defines obesity is the biggest chronic disease people are confronted with,regarded as one of the four largest medical social problem. More than95%of fatpersons are simple obesity who intakes too much energy, leading energy excess,promoting the accumulation of fat, resulting obesity. Recently, people realize that itshould restrict to uptake high fat. However, they think high protein can’t induceobesity, thus, taking in more and more, resulting in obesity.Therefore, the objective of this study is to exact SBTI from soy whey water, toobserve the vitro-stability and apply to the prevention and cure of dietary obesity,providing rationale of development of new losing weight material and a new directionof prevention and cure of obesity.1. Stability of SBTI during digestion in simulating gastrointestinal juice:simulating digestive juice was prepared according to USP. Treated with different time,the degree of hydrolysis of SBTI was determined. The result showed that after treated with simulating gastrointestinal juice, the top degree of hydrolysis was only10%,which indicated that SBTI was stable in simulating gastrointestinal juice. So it couldguess SBTI was stable in vivo.2. Establishing obesity model: weaning ICR mice were randomly divided into2groups. One was normal mice (NM) feeding normal diet, one was fat mice (FM)feeding high fat diet which was made of normal diet adding lard and sucrose to. Thetotal energy of normal diet was3850KJ/KG of which fat providing was11.7%. Thetotal energy of fat diet was4850KJ/KG of which fat providing was48.5%. After6weeks period of modeling, obesity model was established successfully for the bodyweight of FM group was20%heavier than that of NM group.3. The obesity prevention experiment of high fat diet mice: after establishingobesity model, as the obesity prevention group, NM group was divided again into5groups. Except the blank control group, the rest were fed high fat diet instead ofnormal diet, and orally administrated SBTI to observe the effect of prevention. Theresult showed that, compared with control group, weight of groups with SBTIincreased little and triglyceride, high-density lipoprotein and free fatty acid of thatwere improved significantly (P<0.05). So it can say SBTI is effective in preventionof high fat diet mice.4. The reducing weight experiment in high fat-induced obesity mice: afterestablishing obesity model, as the cure experiment, FM group was divided again into5groups. Except model control group, the rest were fed normal diet instead of highfat diet and orally administrated SBTI to observe the effect of reducing weight. Theresult showed that, compared with control group, weight of groups with SBTIdecreased significantly(P<0.05), triglyceride, high-density lipoprotein and free fattyacid of that were improved significantly(P<0.05). So it can say SBTI is effective incure in high fat diet-induced obesity mice.
