As the Chinese economy maintains the higher growth rate,people's living level have improved a lot,and people's requirements for the meat,egg and milk are gradually improving,especially the constantly increasing demand for protein.Soybean is rich in superior protein,and soybean trypsin inhibitor,one of anti-nutrition factors;the latter is bad forpeople's health.Therefore,it is necessary to get rid of the trypsin inhibitor in soybean under mild conditions,through which the nutritional value of soybean can be enhanced,which will have great significance for improving people`s health.In this study,soybean was used as starting material,in which the trypsin inhibitor was eradicated by bio-inactivation technique.A secondary orthogonal regression design of four factors model was constructed to explore the optimal conditions of inactivaty based on the data of inactivity ofsoybean trypsin inhibitor.The methods and result of this study were followed:Exogenous protease technique: Regression model analysis showed that the pH,temperature,time and enzyme dosage affected the removal rate of soybean trypsin inhibitor significantly,which had some interactions.The test results show that the maximal removal rate of soybean trypsin inhibitor could be realized using alkaline protease,with the optimal pH 9.0-9.3,the temperature at 50.2-51.5 ?,the reaction time at 55.1-59.5min,and the amount of enzyme in the range of 923-1048 kU.The verification experimental results are basically consistent with the theoretical values.Endogenous protease technique: Regression model analysis showed that temperature,time,pH,and water quantity during budding had great influence on the removal rate of of soybean trypsin inhibitor,which showed interactions.The experimental results showed that the optimum conditions of endogenous protease method were 22.2-22.6 ?,67.8-78.9 h,pH 6.5-6.7,and the water absorption was 192.2%-214.3%.The experimental results were basically consistent with the theoretical values.The endogenous enzyme method and the exogenous enzyme method are mild which can keep the nutritional value better.The results obtained in this paper provide data support for biological removal of soybean trypsin inhibitors.
