Pentachlorophenol was used as drug to kill the oncomelania and incidental fishes,due to the chemistry of the stability of the past to use pentachlorophenol in the residueenvironment will continue to be quite a long time. The quality of fishery aquatic arestill more serious influence. Industry of Eriocheir sinensis in our country has greatprospects, but the pentachlorophenol was detected many times, to ensure the safety ornot of Eriocheir sinensis were used pentachlorophenol, the detection methods ofpentachlorophenol and eliminate of pentachlorophenol in Chinese mitten-handed crab(Eriocheir sinensis) should be studied, including:(1) Study on the determination ofresidues of pentachlorophenol in fishery aquatic (2) Study on the eliminate ofpentachlorophenol with different doses in Chinese mitten-handed crab, Eriocheirsinensis (3) Study on the existence of pentachlorophenol in different tissues ofChinese mitten-handed crab, Eriocheir sinensis:1: Residues of pentachlorophenol and its sodium salt’s residues in edible tissuesof fishery aquatic were determined with gas chromatography by internal standardmethod. The target compounds were extracted with n-hexane and then back extractedwith potassium carbonate solution. The extract was derivatized with acetic anhydride,and the derivative was extracted with n-hexane. The HP-5MS (30m×250μm×0.25μm) column and the micro-cell electron capture detector was used for thedetermination. This method choose2,4,6-TBP as internal standard substance. Theworking curves were linear (r=0.9996) in the range of0.2~20μg/L. When spiked with2.0,5.0,10.0,50.0μg/kg of pentachlorophenol and50μg/kg of2,4,6-tribromophenol,the average recoveries were85.8%~109.0%and the RSD were0.8%~9.3%. Thedetection limit was0.6μg/kg.2:Residues of pentachlorophenol and its sodium salt’s residues in edible tissuesof fishery aquatic were determined with gas chromatography mass spectrography.The ion source of this method was NCI,the select reaction monitoring was SIM, the current of lamp filament was50μA,the temperature of auxility was250℃,theenergy of electronic was70eV,the temperature of ion source was150℃;thequantify ion of pentachlorophenol was307.8,the qualify ons of pentachlorophenolwere286.8、288.8、305.8、307.8;the quantify ion of pentachlorophenol internalstandard was313.8,the qualify ions of pentachlorophenol internal standard were294.8、296.8、313.8、315.8. The recoveries of pentachlorophenol were87.2%~106.9%, this method was well for detecting of pentachlorophenol residues in aquaticproducts.3. Residues of pentachlorophenol and its sodium salt’s residues in edible tissuesof fishery aquatic were determined with gas chromatography mass spectrographyand mass spectrography. The ion source of this method was NCI, the select reactionmonitoring was SRM, the current of lamp filament was50μA,the temperature ofauxility was250℃,the energy of electronic was70eV,the temperature of ionsource was150℃, the parent ion of pentachlorophen was307.8, the product ions ofpentachlorophen were230.9、265.0; the parent ion of pentachlorophenol internal was313.8, the product ions of internal standard of pentachlorophenol were236.7、271.0.The recoveries of pentachlorophenol were97.5%~104.4%, this method was wellfor detecting of pentachlorophenol residues in aquatic products.4: The eliminate of pentachlorophenol following by different doses ofpentachlorophenol(1000μg/kg,200μg/kg,50μg/kg) with extravenous was carriedout in Chinese mitten-handed crab, Eriocheir sinensis. At nature water temperature,hemolymph, muscle, liver and gill samples were obtained after extravenous inpentachlorophenol solution1days, and then put them in pond to carry eliminateexperiment. The samples were determined by Gas chromatography detection method,the concentration-time data derived from the experiment is processed and thecorresponding pharmacokinetic parameters were calculated with DAS. Theexperiment displayed the pentachlorophenol level in hemolymph, muscle, liver, gillreached maximum momentarily then declined and last tended to stabilize at threeadministration doses. The drug concentration of several tissues from big to small inorder as gill, liver, muscle, hemolymph when processing enrichment experiment. In eliminate experiment, in the extravenous concentration of50μg/kg, the time of theconcentration of pentachlorophenol in liver, gill, hemolymph, muscle under thedetection limit were60days,76days,76days,108days; in the extravenousconcenrration of200μg/kg, the time of the concentration of pentachlorophenol inliver, gill, hemolymph, muscle under the detection limit were124days,124days,108days,156days; in the extravenous concenrration of1000μg/kg, the time of theconcentration of pentachlorophenol in liver, gill, hemolymph, muscle under thedetection limit were all156days. The important parameters were listed. Theabsorption half-life (T1/2α) and the elimination half-lives (T1/2β) in hemolymph were0.748,1.09,3.06d and7.03,7.77,6.02d, respectively, the total body clearance(CLs) was0.0414,0.0641and0.117μg/kg·d, the elimination half-lives (T1/2β) in gillwere3.46d,2.49d,3.93d, the elimination half-lives (T1/2β) in liver were5.63d,5.85d,6.93d and the elimination half-lives (T1/2β) in muscle were5.48d,7.20d,6.42d.5. Through three different extraction methods to research the existencemorphology in the four tissue of Eriocheir sinensis of pentachlorophenol sodium.The research result displayed that their are three different forms: pentachlorophenolpentachlorophenol sodium and the solder with other material, the four organizationof gill、liver、hemolymph and muscle have three different forms, the extractingeffect of extracting method in the process is different, the first extracting method iseasy to column, easy to operated, in order to extract the amount ofpentachlorophenol sodium we choose the first extracting method.
