Haliotis discus hannai Ino is one of important molluslk, it is mainly distributedin the north coasts in china, with tender tastes and mafrional values, it exhibits highvalue and functional components. So far, the abalone was employed in traditionalChinese medicine for protecting the liver, treatment bronchial, and strength kindey. Sofar, very limited documents were reported, and the astilization of abalone exhibiledpromise future. Trough the chemical study of nucleoside components, we testify theliver protection, with aim to explore the role of butanol extracts, and protective effectsin modle of mice. The main results obtained are as follows:1. Isolation and structure determination: in this paper,14compounds wereisolated by extracting Haliotis discus hannai ino gonad with column chromatographyof silica gel, Sephadex LH-20and preparative TLC. Structures of9compounds wereelucidated by means of spectroscopic including TLC、GC-MS、MS、1H-NMR、13C-NMR、1H-1H-COSY and analysis of their physical-chemical characters. Theirstructures were as: methyl pyroglutamate(1),1-methyl thymine(2),2’-deoxyadenosine(3), uracil(4),2’-deoxyuridine(5),3-Hydroxy-4-hydroxymethyl-4-butanolides(6), uridine(7),3-methyl thymine(8),1,3–propanediol(9). All the compounds were isolated from this marine mollusk forthe first time.2. Studies on the protective effects and mechanisms of butanol extraction fromthe Haliotis discus hannai Ino against acute chemical liver injuries in mice:(1)CCl4-induced hepatotoxicity was manifested. Pretreatment of mice the butanolextraction from the Haliotis discus hannai Ino, significantly reduced the levels ofserum enzyme ALT、AST on acute chemical liver injury which were induced by CCl4. The liver MDA was decreased and the activites of SOD was increased.(2)D-GalN-induced hepatotoxicity was manifested. Pretreatment of mice the butanolextraction from the Haliotis discus hannai Ino, significantly reduced the levels ofserum enzyme ALT、AST on acute chemical liver injury which were induced byD-Gal. The liver MDA was decreased and the activites of SOD was increased.(3)Thecontents and results were dose-effect relationship. The levels of ALT and AST andcontents of MDA will be smaller, activities of SOD will be much more higher, whencontents were more.
