The Paris polyphylla(chonglou) is the dried rhizome of Pris polyphylla Smith var.yunnanensis or Pris polyphylla Smith var.Chniensis. It has played an important rolein traditional medicine development for anti-tumors,stanch bleeding,immunityadjustment,analgesia and so on.The active components are a series of steroidglycosides. An accurate method for determination of the content of total saponinscan be used for quality control of Paris polyphylla. The traditional techniques ofsolvent extraction of plant materials are mostly used of methanol and ethanol.Applying biological enzyme to catalyze the efficient components to release andextract. A technology for extracting total saponins from Paris polyphylla was studiedby using cellulolytic enzyme treatment and organic solvent extraction methods.The main results contains as follows:First of all, a quantitative analytical method of total saponins was studied bycolorimetry. The samples analyzed were first reacted with the solution of5%vanillin in acetic acid and70%perchloric acid(4:6) at50℃for25min. Then, thecontents of total saponins in Paris polyphylla samples were determined byspectrophotometry at457nm. The calibration curve was linear in range of0.02~0.14mg. The average recovery and relative standard deviation were96.60%and3.41%respectively. The method is simple, accurate and reproducible and can beused for quality control of Paris polyphylla.Then, in order to increase the yield of total saponins, the technology ofultrasonic-assisted enzymatic method were used. Base on the single factorexperiments and the Box-Behnken center united experimental design principle,effects of enzyme concentration, enzymolysis temperature, pH value andenzymolysis time on extraction yield of total saponins were investigated. The resultshowed that the optimum extraction condition was obtained as follows: enzymeconcentration32U/g, enzymolysis temperature52℃, pH value4.6, enzymolysistime92min. The predictive maximum yield of total saponins is1.68%.The averagevalidation value is1.66%.The relative error between them is1.12%.Finally, the results of antioxidant activities showed that scavenging activity ofthe extract for hydroxyl radical and superoxide anion was favorable, the half-scavenging concentration (IC50) were0.4mg/mL and0.6mg/mL, respectively. Theresults also showed that total saponins had remarkable antioxidant activities in vitro....
