Salidroside is an active component of Chinese herbal medicine Rhodiaola crenulata(Hongjingtian in Chinese). Fluorescence properties of salidroside and Rhodiaola crenulatawere studied in this thesis, and fluorometric method and thin-layer chromatography (TLC)were proposed for the determination of salidroside in Rhodiaola crenulata. Fluorescenceproperties of Rhizoma Zingiberis, Ginger,6-gingerol who is an active component ofZingiberis and Ginger, and Cimifugin were also studied in the thesis.Three parts were included in the thesis:1. Fluorescence property of salidroside was studied. The influence of acidity, surfactantand solvent on fluorescence property and the linear relation between fluorescence intensityand solution concentration were investigated. Using L-tryptophan as a reference and in nearneutral condition, fluorescence quantum yield of salidroside was measured to be0.156atmaximum excitation wavelength279nm. The fluorescent component of Rhodiaola crenulatawas studied by TLC method, and the result revealed that only salidroside is the fluorescentcomponent. Based on this fact, a solution fluorometric method was proposed for thedetermination of salidroside in Rhodiaola crenulata medicinal crops, and the standard additionmethod and the standard curve method were used respectively to determine the content ofSalidroside in Rhodiaola crenulata, the results are basically the same. with2. Fluorescence properties of Rhizoma Zingiberis,Ginger,6-gingerol were studied. Theinfluence of acidity, surfactant and solvent on fluorescence property was investigated.Three-dimensional (3D) fluorescence fingerprints of Rhizoma zingiberis and Ginger werestudied, and a3D fluorescence examination method for identification of Rhizoma zingiberisand Ginger was established.3. Fluorescence properties of cimifugin were studied. The influence of acidity, surfactantand solvent on fluorescent property was investigated. Using L-tryptophan as a reference andin pH9.22, fluorescence quantum yield of cimifugin was measured to be0.023at maximumexcitation wavelength296nm.
