In orde to reduce the influence caused by algae bloom, lots of studys were focused on the cyanobacterial prevention in the large lakes worldwide, but few works focused on the small rural surface water. To investigate the microcystin-producing cyanobacteria in rural surface water samples, different types of rural surface water were sampled from Huizhou, including pool water, reservoir water and running water. Molecular techniques and HPLC were employed to detect the microcystin-producing cyanobacteria and MCs. The results showed that mcyA-Cd, the specific fragment coding for the microcystin synthetase complex, can be used to distinguish the toxic cyanobacterial and non-toxic cyanobacterial effectively. PCR-DGGE in combination with cloning and sequencing proved that the primary detection of microcystin-producing cyanobacteria in the environmental sample using mcyA-Cd is of high specificity and accuracy.Study has nosed out the eutriphication in the rural surface water samples from Huizhou was serious. The concentration of TN and TP in most of the water samles exceeded the guidline which gived by GB3838-2002. The water resources of the sampling sites have been contaminated by microcystin-producing cyanobacteria., most of the microcystin-producing cyanobacterias were Microcystis. It was also observed that TP concentration showed significant positive correlations with concentrations of MC-producing cyanobacteria, but the correlation between TN and MC-producing cyanobacteria was not significant. HPLC analysis indicated that MC-RR or MC-LR was detected in samples, even in the pool water and running water. Most of the samples have been contaminated by MC-RR. These results suggest that appropriate measures should be taken to monitor the microcystin-producing cyanobacteria in the rural surface water in Huizhou, in order to avoid or reduce the potential hazards to human. The results also helped to give a assessment for the water quality.
