Nature clay minerals, such as montmorillonite (MMT) and attapulgite (At) can be used as pharmaceutical excipients, which have good prospects for drug delivery and control release. But there are little further development of clay in drug or therepy method.First of all, This paper focused on the intercalation of polyethylenamine (PEI) modified Doxorubicin/MMT as a gene and drug carrier.Secondly, based on the above, the paper compared the physical, chemical and cytology properties of the different molecular weight PEI/5-fluorouracil/montmorillonite, and found the lower molecular weight PEI/5-fluorouracil/montmorillonite was better in transfection efficiency.Finally, thinking of the characteristics of oral medication, PEI1200modified montmorillonite and attapulgite was prepared, which was load5-fluorouracil (5-FU) or capecitabine (Cap), and condensed pTRAIL for early gastric cancer cell apoptosis-inducing, we also proved the PEI/MMT is available for the protection of the gastric mucosa.the good performance of PEI/drug/clay system particle size, decreased, drug release profile, cytotoxicity and gene transfection efficiency. The co-delivery of anti-cancer drug and pTRAIL plasmid induced apoptosis of gastric cancer cell lines effectly, and It revealed gastric mucosa damage protectation from alcohol by PEI/MMT.
