Dagao enzyme is a kind of product that fermentation with a special process, with which it needs a180days of natural fermentation with more than50kinds of plants such as fresh fruits, vegetables, fungusmushroom, and so on. Without dilution, people could use it directly. It is rich in nutritions such as vitamins,minerals, amino acids, organic acids, oligosaccharides, and so on and plays a role in active cells、agingresistance、improving health. It mainly refers to the methods on《Technical standards for testing andassessment of Health food》(2003edition) to study the safety and function of the functional food--Dagaoenzyme.Firstly, the safety of Dagao enzyme was evaluated with actue toxicity test and subchronic toxicity test.The actue toxicity test was conducted with mice though determinate the lethal dosage50and maximumtolerated dose. The results showed that there was no death of mice and not get lethal dosage50, and themaximum tolerated dose up to108ml/(kg·BW) which was equivalent to108times of the recommended oraldoses60ml/(peraonal·day). The subchronic toxicity test was conducted with SD rats, after90days continuousgavage with Dagao enzyme, the general behavior, the function of liver and kidney and pathological changeswere analysised. The results showed that the rats were in a good condition, and there was also no symptom ofpoisoning, no death and no difference in indexes of organs and histopathologic examination. It had no toxicrole in liver and kidney of rat and played certain protective effect on the liver.Secondly, constipated mice models were established. Through the test of small intestine movement anddefecation of mice, the effect of relieving constipation was studied and the outcome showed that Dagaoenzyme improved the propulsive motility of small intestine of mice significantly and increased the volume aswell as weight of feces with earlier first defecation time. To study the mechanism of enhanced body immune,the ConA-induced lymphocyte transformation and natural killer (NK) cell activity was determined and theresults showed that Dagao enzyme could significantly improve the proliferation of T cells but it could notimprove the activity of NK cell.At last, to discuss the function of Dagao enzyme in antialcoholism and liver protection, the model ofacute liver injury induced by alcohol in mice was established and the contents of ALT, AST, TG, ADH, MDAin the liver tissue were detected. The results showed that Dagao enzyme could significantly lower the enzymeactivity of AST and ALT in liver tissue, reduce the injury of alcoholic on liver, improve the activity of ADHin liver tissue, promote alcohol metabolism and decrease alcohol concentration.The results above indicate that Dagao enzyme is a kind of safe, nontoxic, health beverage, and it has agood performance in antialcoholism and certain effect in protecting liver, relieving constipation, improvingcellular immunity.
