Liver-protection of Ge Gan Capsule is a health food which has auxiliary protective effects on chemical liver injury with radix puerariae,radix glycyrrhizae, fructus schisandrae, rhizoma curcumae longae as raw material.In this paper, first of all, radix puerariae, radix glycyrrhizae were identified according to "national pharmacopoeia of the People’s Republic of China(first)”.Extraction process: first of all, utilizing the single factor experiment to inspect the concentration of ethanol, the extraction times, the extraction time, the liquid-solid ratio.On the basis of single factor experiment, with ethanol concentration, extraction time,extraction times and liquid-solid ratio as the impact factor, with puerarin extraction rate and glycyrrhizic acid extraction rate as the response value, the optimized combination condition is obtained by response surface analysis. The experimental results show that regression equation of the curved surface nicely fitted with the experimental results and the optimized combination condition is adding 15 times amount of 64.50%ethanol technology for medicinal materials, extracting3 times, 3h each time. Under the optimized combination condition, puerarin extraction rate and glycyrrhizic acid extraction rate predicted is 81.00%、86.62%.The validation test showed puerarin extraction rate is83.00% and glycyrrhizic acid extraction rate is 87.02%.The formation techniques were studied. Liver-protection of Ge Gan capsule is a health food that has auxiliary protective effect on chemical liver injury and based on dry powder which used the radix puerariae, radix glycyrrhizae, fructus schisandrae, rhizoma curcumae longae extracted by the optimized combination condition and the right amount of starch. Investigation into the wetting agents, the drying time,the difference of content,the time of disintegration, process routing of experiment is stable, simple, the item of check is qualified.suitable for industrial production.The quality standard: the effective components of radix puerariae,radix glycyrrhizae,fructus schisandrae, rhizoma curcumae longae were determined with TLC, UV and HPLC was used to determine the content of total flavonoids and puerarin.Result: the spots were clear and well separated. the content of total flavonoids in Liver-protection of Ge Gan Capsule was 32.95 mg/g, The average content of puerarin in Liver-protection of Ge Gan Capsule was 47.0 mg/g.Functional experiment :This paper also studied the Liver-protection of Ge Gan Capsule the protection. function on carbon tetrachloride(CCl4) in rats with acute liverinjury and alcoholic liver injury. Results: Liver-protection of Ge Gan Capsule has auxiliary protection function on chemical liver injury and reduce ALT and AST caused by CCl4.At the same time, it can reduce liver damage and liver steatosis and liver steatosis. And then it was verified protective effects on alcoholic liver injury, and has the potential to prevent fatty liver function.
