Hu Luba, the dry and mature seeds of the Trigonella foenum-graecum L.(Fenugreek seeds),which mainly distributed in Henan, Anhui, SichuanProvince. Its taste is bitter, sex is warm, attributable to kidney meridian.Fenugreek seeds have the functions of warming kidney, dispelling pain andect. Fenugreek seeds contain the chemical composition of flavonoids,alkaloids, saponins and ect.Modern pharmacological research show that,vitexin has the effect of anti-inflammatory, anti-bacterial, anti-spasmodic,dilat-ion of blood vessels and ect;trigonelline has significant effect of loweringblood sugar, reduce cholesterol, sedation and other physiologicalactivity.Therefore,this topic studyed the Hu Luba extraction preparationprocess with the vitexin and trigonelline extraction rate as the evaluationindex. In order to effectively control the quality of Hu Luba extraction,theTLC identification and content determination experiments were researchedand has drawn up the quality specification draft. The different batches of HuLuba extraction HPLC fingerprint was established. Expecting to provide ascientific basis for the control of the quality of Hu Luba extraction.In order to obtain stable quality, high active ingredient content of the HuLuba extraction,this paper conducted the extraction preparation craftresearch.In the research, L9(34) orthogonal was designed to optimizedextraction process based on the physical and chemical properties of vitexinand trigonelline. Through investigate the ethanol concentration, extraction time, liquid ratio and extraction times, determined the best extract process.Validation test results show that the technology was stable and feasible.In order to quickly and effectively identify Hu Luba extraction, the thinlayer chromatography has carried on as the qualitative identification whichthe reference substance are vitexin and trigonelline. By investigating thepreparation of sample solution, selection of expansion agent and theproportion of adjustment, established the Hu Luba extraction qualitativeidentification method. Experimental results show that the method operation issimple, and spot color clear, than move value is moderate, with goodreproducibility.In order to be able to accurately reflect the quality of Hu Luba extraction,its extractive was determined by HPLC,using the vitexin and trigonelline asthe reference substance.The extraction solvent, extraction and preparationmethods of the test time has been studyed during the sample preparationprocess. The chromatographic column, mobile phase and detectionwavelength was investigated, and the chromatographic conditions wasdetermined for the determination of the content. Methodology inspection testresults show that the content determination method has high accuracy, goodreproducibility, can be used as a quantitative detection extracts characteristiccomposition method.To overall evaluation about the quality of the Hu Luba extraction, thisarticle carries on the experimental research of the fingerprint. In the test,mobile phase gradient elution conditions, the best absorption wavelength andthe detection time were investigated, and the optimum solution wasdetermined. Vitexin as reference, the10batches of Hu Luba extractionfingerprint of flavonoids ingredients was established.With reference to the Chinese Pharmacopoeia, the character, identification,moisture content, heavy metals content and on ignition residue and content determination had been provided.Preliminary draft of quality standard wasworked out,which laied the foundation for the establishment of Hu Lubaextraction quality standard system and provided a reliable scientific basis toeffectively control the immanent quality of Hu Luba extraction.
