Root is an important organ of plants, and play the important role of fixing and absorbing nutrients. Root cap is located in front of the root, in addition to protecting the root tip meristem, it is the first organ to feel and pass the signal from underground. The structure and development of root cap have been very clear in dicotyledonous plant Arabidopsis thaliana. Comparison with Arabidopsis, related research has lagged behind in rice. Therefore, knowing the mechanism of root development will play a very important role in building a more complete regulatory networks for root development and modifying rice. Compared with Arabidopsis, the research for rice root cap is more difficult. As the root cap is easy to fall off, and more thick than the Arabidopsis. It is difficult to see under the microscope, the method of slice must be used. Studies on the development of rice root cap just begin, there is no specific marker. The number of layers of rice root cap cells is unknown? It is not clear how the rice root cap development?We used the approach of forward genetics and reverse genetics to study the rice root system, the main study results were described as follows:1. After the seed sterilization and planting the seed in a vertical way, I screened T-DNA insertion mutant library, and found more than 70 markers expressed in the rice root cap, got four flanking sequences.2. Through bioinformatics approach, I found the rice root cap development gene Os02g15340 and Os06g33940, and successfully constructed promoter fusion vector, overexpressed vector, amiRNAs vector, complementary vector and transformation.3. Through resin sections, we have knowed the structure of rice root cap, which is about 13-14 layers of cells, and the possible development process of rice root cap.4. After treating rice root cap by auxin, cytokinins and other six kinds of hormones, we found that NAA,2,4-D and ABA affect the morphology of root cap, other hormones had no effect on the root cap, and layed the foundation for rice root development.
