Mei flower(Prunus mume Sieb.et Zucc.)is a traditional famous tree of China, which was widely used in landscape architecture.Nowadays, the studies on Mei flower involves germplasm resource, cultivation, tissue culture and molecular biology and so on. The molecular marker is widely used in cultivar classification of Mei flower, but no article about construction of genetic linkage maps in Mei flower was reported.A frame genetic map of Mei flower was constructed with AFLP and SSR markers using F1 population which was derived from’Fenpi Gongfen’×’Xuemei’.The main results are described below:1.Construction of mapping populationThe F1 popualtion contained 56 individuals, which were derived from hybridization between’Fenpi Gongfen’and’Xue Mei’.2.AFLP polymorphism analysis24 primer pairs with high polymorphism and clear bands were selected from 512 AFLP primer combinations.By primers screening for analysis of F1 population,167 polymorphic loci were obtained, of which,51 ones in’Fenpi Gongfen’,56 in’Xuemei’, 73 appeared in both parents.3.Sourse of SSR primers89 primer pairs were downloaded from genome database of rosaceae (http://www.bioinfo.wsu.edu/gdr/,GDR), most of these primers are relative species of Mei flower in same genera, like almond and peach. In the same time, using combination method of FIASCO method and magnetic beads enrichment,52 primer pairs were designed and synthesized, which contain 50 pairs produced AG repeats and two pairs generated CAG repeats.4. SSR polymorphism analysis17 primer pairs of polmyoprhism, high repeatability and clear bands were screened for polymorphism analysis,12 polymorphic loci were obtained,4 in’Fenpi Gongfen’,5 in’Xuemei’,3 appeared in both parents.5.Construction of frame genetic linkage mapUsing JOINMAP3.0 for linkage analysis,linkage map was constructed separately in the two parents.The map of’Fenpi Gongfen’was composed of 42 markers, distributed in 14 linkage groups, which covers 305 cM of mei flower genome. The length of map of ’Xuemei’is 232cM, including 48 markers which were distributed in 14 linkage groups.
