Peppers, Capsicum (Family: Solanaceae), is originated in Latin America tropicsand has been introduced in China for many years. As with other crops, vulnerability ofthe pepper genotypes to a multitude of abiotic and biotic stresses has restricted theirpotential yield. Biotechnology techniques of plant regeneration in vitro and genetictransformation are powerful tools can propel the Capsicum improvement andcomplement conventional breeding. But the efficiencies described in the literature forpepper regeneration or transformation lag behind other solanaceous crops due to itsrecalcitrant natural and high genotypic dependence, which in turn makes it difficult toapply recombinant DNA technologies aimed at genetic improvement against pests,diseases or abiotic stress.Capsicum annuum var. annuum (cv. Xinsu) was used as our experimental materials.Based on the uniform design and the mathematical model equation, we eatablish aeffiective regeneration in vitro.The results showed that cytoledon was the best type ofexplant and12-day-old was the best choice of seedling age; MS+IAA5.71μM+BA22.20μM+AgNO329.43μM was effective for shoot bud induction with thefrequency of99.20%; MS+IAA5.71μM+BA4.44μM+AgNO329.43μM+GA35.77μM was most suitable for plant elongation with the rate of86.67%; MS+0.57μMIAA+0.69μM NAA was effective for plant rooting with the ratio of98.33%. Thesystem of plant regeneration of pepper was established successfully.COI1gene encodes a F-box protein to assemble SCFCOI1complexes that wereessential for reponses to jasmonate(JA). COI1gene regulates plant pollen fertility,defense against pathogens and insects. Based on the PBIN19,the RNAi vector wasestablished. The transgenic pepper were developed by Agrobacterium-mediated planttransformation with Km resistance selection. The results showed that silence transgenicpepper are shorter than wide type and the resistance is more weaker.
