Effect Of Vitamin E On Performance,antioxidant,lipid Metabolism And OBR Gene Expression Of Breeding-hens

The trial was designed to observe the effect of diet supplemented with different levels of Vitamin E on Performance,Egg quality ,Meat quality ,Antioxidant enzyme activity,Alpha-tocopherol in the egg yolk and tissue,Lipid metabolism and OBR gene expression,and discussed the role of mechanism which could provide the oretical basis for the application of VE on production of Breeding-hens.At the age of 48 weeks, 264 Hy-line were randomly divided into 6 groups, with 4 replications per group and 11 hens per replication. The control group fed with basal diets, the testing group fed with basal diets adding 20mg/kg, 40mg/kg, 80mg/kg, 160mg/kg, 320mg/kg Vitamin E respe- ctively. The whole period consisted of a one week pretest and a eight weeks test.The result showed that:①The vitamin E supplementation to the diet had an effect on feed intake, GroupⅥ(320mg/kg) significantly decreased the control group (P<0.01),there was no difference among other groups (P>0.05); Average egg weight, egg production rate, the ratio of feed to egg, but the difference were not significant(P>0.05).②Diets supplemented with Vitamin E, in conventional egg quality:egg weight, egg shape index, Haugh unit, relative eggshell weight, relative yolk weight were no significant differences (P> 0.05). Eggshell thickness, group V was significantly different from groupⅢ(P<0.01), the groupⅤwas significantly different from groupⅡand group IV (P<0.05), the first groupⅡeggshell thickness.The groupⅣincreased by 4.35% than the control group.Yolk color, the groupⅢand group IV were significantly different from groupⅡ( P <0.01), the groupⅢand group IV were different from the group, groupⅤand groupⅥ(P> 0.05), the group III has the deepest color than other groups,It increased by 6.67% than the control group.The Yolk cholesterol of groupⅥwas significantly lower than control group (P<0.01), groupⅡa nd groupⅤwere lower than the control group (P<0.05), groupⅡand groupⅢwere no differences with control group (P<0.05),The difference between the experimental groups was not significant (P>0.05). ③Diets supplemented with Vitamin E, in the pectoral muscles, drip loss: group III, group IV and group V were significantly lower than the control group (P<0.01), there was significant difference between group VI and the control group, groupⅡ(P<0.05); 45-minute pH: group VI was significantly higher than the control group (P<0.05); 24-hour pH: group VI significantly higher than the control group (P<0.01), significantly higher than the groupⅡ(P<0.05); The shear force of each experimental group a decreased trend compared with control group, but the difference was not significant (P>0.05).In Leg muscle,drip loss: group III was significantly lower than the control group (P <0.01); within 45-minute pH: groupⅤs ignificantly higher than the control group and groupⅡ(P<0.01), group III, IV, VI was significantly higher than the groupⅠ,group II (P<0.05); 24-hour pH: there was an increased trend among groups,but the difference was not significant (P>0.05); shear stress: group IV and groupVI was significantly lower than the control group (P<0.01),groupⅢwas significantly lower than the control group (P<0.05); Group IV and VI was significantly lower than group II (P<0.05), and the difference was not significant among other groups (P>0.05).④D iets supplemented with Vitamin E, in the serum , SOD: the experimental groups was significantly higher than the control group (P<0.01) , group VI was significantly higher than the groupⅠ,group II, group III and group IV (P<0.01), group VI was significantly higher than group V (P<0.05) ; GSH-PX: control group was significantly lower than other experimental groups (P<0.01), groupⅥwas very significantly higher than the groupⅠ, groupⅡ, groupⅢand group IV (P<0.01), significantly higher than groupV(P<0.05), and the difference was not significant among groupⅢ, groupⅣand groupⅤ(P>0.05). MDA: the experimental groups were significantly lower than the control group (P<0.05).⑤D iets supplemented with Vitamin E, in the liver , SOD: experimental groups were significantly higher than that of the control group (P<0.01), groupⅤ, groupⅥwere significantly higher than the group II, group III and group IV (P<0.01), the group VI significantly higher than groupⅤ(P<0.05); and the difference are not significant among group II, group III and group IV (P>0.05),Group VI increased by 64.85% than the control group; GSH-PX: the experimental groups were significantly higher than the control group (P<0.01), group III,groupⅡ, groupⅤand group VI were significantly higher than groupⅡ(P<0.01), the difference was not significant between group III and group IV (P>0.05) ,and the difference was not significant between groupⅤand groupⅥ(P>0.05); MDA: groupⅥwas very significantly lower than theⅠ,Ⅱ,Ⅲand groupⅡ(P<0.01), groupⅤwas significantly higher than the groupⅠand group II (P<0.05), and there was no significant difference between groupⅤ,groupV Iand groupⅡ(P> 0.05) ,Group VI decreased by 31.51% than the control group.⑥D iets supplemented with Vitamin E, in the pectoral muscle, both SOD and GSH-PX : groupⅥsignificantly higher than the groupⅠ,group II and group III (P<0.01).The difference was not significant between groupⅥand group IV, group V (P>0.05), and the remaining groups had no significant difference but groupⅥ(P>0.05); MDA: groupⅥw as significantly lower than that of the control group (P<0.01). Group II, group III, group IV, and groupⅤwere significantly lower than groupⅠ(P<0.05).The difference among the experimental groups were not significant (P>0.05).⑦Diets supplemented with Vitamin E, in serum ,total cholesterol:Group III, IV and groupⅤw ere significantly lower than that of the control group (P<0.01), and groupVI was significantly lower than groupⅠ(P<0.05), the difference between groupⅡa nd the control group was not significant (P>0.05) , the difference was not significant among the experimental groups(P>0.05); triglycerides: group III was significantly lower than the control group (P<0.05), there was no significant difference among the other groups (P>0.05); low-density lipoprotein cholesterol: the difference was not significant (P>0.05); high-density lipoprotein cholesterol: GroupⅡ,Ⅴand group VI were significantly higher than the control group(P<0.01), groupⅤa nd groupⅥwas extremely significantly higher than that groupⅡ(P<0.01), group III was significantly higher than the control group(P<0.05), the difference between group II and control group was not significant (P> 0.05).⑧D iets supplemented with Vitamin E,α-tocopherol deposition in egg yolk: group III, IV,Ⅴand groupⅥwere significantly higher than the control group (P<0.01), and groupIV, VandVI were significantly higher than the groupⅡ(P<0.01), groupⅤand groupⅥwere very significantly higher than groupⅢ( P<0.01);The groupⅥincreased by 409.05% than the control group.α-tocopherol deposition in cardiac: GroupⅡ,Ⅴa nd VI were significantly higher than the group I, II and group III (P<0.01), the difference between groupⅤa nd groupⅥw as significant (P <0.01) .The groupⅥincreased by 151.62% than the control group;α-tocopherol deposition in liver: the experimental groups were significantly higher than the control group (P<0.01) ,there were significant difference among the experimental groups(P<0.01);α-tocopherol deposition in kidney : each experimental group was significantly higher than the control group (P<0.01),the difference among the experimental groups were not significant (P>0.05).⑨D iets supplemented with Vitamin E, There was no significant effect on the OBR gene expression. There was no significant difference among groups (P> 0.05).
